摘要
目的:探讨刺梨提取物对D-半乳糖(D-Gal)所致小鼠急性肝损伤的缓解效能。方法:建立D-Gal所致的急性化学性肝损伤动物模型,通过测定小鼠肝脏指数,检测血清中谷丙转氨酶、甘油三酯以及谷草转氨酶水平,并结合肝脏样本中丙二醛和过氧化物歧化酶活性的评估,全面评价肝功能损伤程度。同时,检测小鼠血清中的过氧化物歧化酶与谷胱甘肽过氧化物酶水平,并观察肝脏病理切片,以此评价刺梨提取物对急性化学性肝损伤的防护效果。结果:刺梨提取物可显著降低小鼠血清中甘油三酯、谷草转氨酶和谷丙转氨酶水平(P<0.05,P<0.01),提高肝脏中过氧化物歧化酶与谷胱甘肽过氧化物酶活力(P<0.05,P<0.01),降低丙二醛水平(P<0.05,P<0.01),同时改善急性肝损伤小鼠肝细胞坏死、肝组织结构紊乱及炎性细胞浸润现象。结论:刺梨提取物可减轻D-Gal诱导的小鼠急性化学性肝损伤,对小鼠肝脏具有一定的保护作用,为刺梨在医药和功能食品领域的开发应用奠定了理论基础。
Objective:To investigate the alleviating effect ofRosaroxburghiiTratt extract on acute liver injury induced by D-galactose(D-Gal)in mice.Method:An animal model of acute chemical liver injury caused by D-Gal was established.The liver index of mice was measured,and the levels of alanine aminotransferase,triglycerides,and aspartate aminotransferase in serum were detected.Combined with the evaluation of malondialdehyde and peroxidase activity in liver samples,the degree of liver function damage was comprehensively evaluated.At the same time,the levels of superoxide dismutase and glutathione peroxidase in mouse serum were detected,and liver pathological sections were observed to evaluate the protective effect ofRosaroxburghiiTratt extract on acute chemical liver injury.Result:The extract of prickly pear can significantly reduce the levels of triglycerides,aspartate aminotransferase,and alanine aminotransferase in mouse serum(P<0.05,P<0.01),increase the activity of peroxidase and glutathione peroxidase in the liver(P<0.05,P<0.01),and reduce the level of malondialdehyde(P<0.05,P<0.01);simultaneously improve liver cell necrosis,liver tissue structural disorder,and inflammatory cell infiltration in mice with acute liver injury.Conclusion:Extracts of prickly pear can alleviate acute chemical liver injury induced by D-Gal in mice and have a certain protective effect on the liver,laying a theoretical foundation for the development and application ofRosaroxburghiiTratt in the fields of medicine and functional food.
