摘要
狼疮性肾炎(LN)是继发性肾小球肾炎,是系统性红斑狼疮(SLE)累及肾脏的严重并发症,迄今发病机制尚未完全明确,可能与免疫功能异常、遗传因素及病毒感染有关。LN治疗周期长,药物毒不良反应明显,复发率高,目前亟待研究中医药对其治疗效果,以期提高LN患者生命质量。Janus激酶/信号转导子和转录激活子(JAK/STAT)信号通路在LN疾病进展中具有关键作用,磷酸化的STAT信号表达可能与LN发病机制密切相关。本文以JAK/STAT信号通路为切入点,总结其在炎症反应、免疫应答、细胞凋亡等方面对LN的调节作用,综合近年来中药单体及其化合物和中药制剂调控JAK/STAT信号通路治疗LN机制研究,认为中药调控JAK/STAT信号通路对LN防治具有重大意义,以期为LN的防治及药物研发提供参考。
Lupus nephritis(LN)is a secondary glomerulonephritis and a serious complication of systemic lupus erythematosus(SLE)affecting the kidneys.The pathogenesis of LN is not yet fully understood,and may be related to immune dysfunction,genetic factors,and viral infections.LN has a long treatment cycle,significant drug toxicity and side effects,and a high recurrence rate.Currently,there is an urgent need to study the therapeutic effect of traditional Chinese medicine on LN patients in order to improve their quality of life.The Janus kinase/signal transducer and activator of transcription(JAK/STAT)signaling pathway plays a crucial role in the progression of LN disease,and phosphorylated STAT signal expression may be closely related to the pathogenesis of LN.This paper takes the JAK/STAT signaling pathway as the entry point,summarizes its regulatory effects on LN in inflammatory responses,immune responses,apoptosis,and other aspects,and synthesizes recent research on the mechanisms of traditional Chinese medicine monomers,their compounds,and traditional Chinese medicine preparations in regulating the JAK/STAT signaling pathway for LN treatment.It is concluded that the regulation of the JAK/STAT signaling pathway by traditional Chinese medicine holds significant importance for the prevention and treatment of LN,with the aim of providing references for LN prevention,treatment,and drug development.
