基于AHP-CRITIC结合Box-Behnken响应面法研制骆驼蓬烟熏剂

Development of Luotuopeng (Peganum harmala L) Fumigant Based on AHP-CRITIC Combined with Box-Behnken Response Surface

摘要 目的:基于主观赋值法(AHP)-客观赋值法(CRITIC)结合Box-Behnken响应面法,以骆驼蓬为主要原料,其他植物粉末为辅料研制骆驼蓬烟熏剂。方法:试验以烟熏剂的体外抑菌活性、骆驼蓬碱、去氢骆驼蓬碱的含量与烟熏剂的外观为指标,对烟熏剂的配方进行优化。采用AHP-CRITIC混合加权法确定各指标的权重系数,联合单因素试验和Box-Behnken响应面法对骆驼蓬烟熏剂配方进行优化。应用GC-MS技术对烟熏剂烟雾成份进行分析,用急性吸入毒性试验评价其安全性。结果:优化后的最佳配方为以骆驼蓬含量为基准,茶粕粉添加量为10.4%、植物助燃剂添加量为40.0%、植物粘粉添加量为8.0%。骆驼蓬烟熏剂对金黄色葡萄球菌、停乳链球菌抑制圈直径分别是19.95 mm、10.52 mm。含量测定结果表明,骆驼蓬烟熏剂中骆驼蓬碱含量为0.88 mg/g,去氢骆驼蓬碱含量为1.30 mg/g。GC-MS共鉴定烟熏剂烟雾105种成份,主要为吲哚类、酚类和脂肪酸类物质,其中去氢骆驼蓬碱在烟熏剂烟雾中相对含量为3.14%。SD大鼠急性吸入毒性试验结果表明,烟熏剂对受试大鼠的急性吸入LC_(50)>101666.7 mg/m^(3)。结论:骆驼蓬抑菌烟熏剂工艺稳定可行、烟熏剂有效安全、生态环保。 Objective:To develop and optimize a Luotuopeng(Peganum harmala L)-based fumigant using Luotuopeng(Peganum harmala L)as the primary raw material supplemented with other plant powders,through an integrated approach combining subjective weighting(AHP)and objective weighting(CRITIC)methods with Box-Behnken response surface methodology.Methods:The test was carried out to optimize the formulation of the fumigant using the antibacterial activity of the fumigant,the contents of Vasicin and Harmine,and the appearance of the fumigant as the indexes.The weighting coefficients of each index were determined by AHP-CRITIC hybrid weighting method,and the formulations of the fumigants were optimized by combined one-way test and Box-Behnken response surface method,and the smoke components of the fumigants were analyzed by GC-MS.The safety was assessed using an acute inhalation toxicity test.Results:With a 10.4%addition of tea meal powder,40.0%addition of plant combustion aid,and 8.0%addition of plant stickiness powder,the optimal formulation was based on Peganum harmala L content.The diameters of the inhibition circles of Staphylococcus aureus and Streptococcus dysgalactiae subsp.dysgalactiae were 19.95 mm and 10.52 mm,respectively,and the results of the content measurements showed that the content of Vasicin in the fumigant was 0.88 mg/g,the content of Harmine was 1.3 mg/g,and the content of Harmine was 1.30 mg/g.A total of 105 components were analyzed in the smoke of the fumigant,mainly indoles,phenols and fatty acids,and the relative content of Harmine in the smoke of the fumigant was 3.14%.The results of acute inhalation toxicity test in SD rats showed that the acute inhalation LC_(50) of the fumigant to the subject rats was>101666.7 mg/m^(3).Conclusion:It indicates that Peganum harmala L bacteriostatic fumigant process is stable and feasible,effective and safe,ecological and environmentally friendly.
