摘要
试验旨在探究怀乡鸡肠道中分离的4株卷曲乳杆菌的益生特性。采用FITC荧光标记结合分光光度法测定菌株对Caco-2细胞的黏附率,依靠不同pH值的生理盐水及不同含量的胆盐MRS培养基评估菌株耐酸耐胆盐能力,利用人工胃液和人工肠液分析菌株的消化液耐受性,采用分光光度法测定菌株自聚集能力,二甲苯分离法评估菌株疏水性,基于ResFinder和毒力因子数据库(VFDB)数据库注释菌株功能基因。结果显示,所有菌株对Caco-2细胞的黏附率均高于25%,在pH值3.0和0.1%胆盐的环境下均具有良好耐受性,在人工肠液中存活率较高。菌株自聚集能力强,疏水性均高于55%,这可能与菌株表面S层结构相关。基因功能注释结果显示,菌株携带多种益生相关功能基因,具备较强的环境适应性和安全性。研究表明,4株卷曲乳杆菌均具有良好的胃肠道适应性与定植能力,可作为家禽益生菌的候选菌株。
The experiment aimed to investigate the probiotic properties of four Lactobacillus crispatus strains isolated from the intestinal tracts of Huaixiang chickens.Adhesion to Caco-2 cells was quantified using FITC fluorescence labeling and spectrophotometry.Acid and bile salt tolerance were assessed using saline at varying pH values and MRS media supplemented with different bile salt concentrations.Resistance to simulated gastrointestinal fluids was evaluated using artificial gastric and intestinal juices.The auto-aggregation capacity was measured by spectrophotometrically,while cell surface hydrophobicity was assessed via xylene partitioning.Functional genes were annotated based on the ResFinder and Virulence Factors of Pathogenic Bacteria(VFDB)databases.The results showed that the adhesion rate of all strains to Caco-2 cells was higher than 25%,and they were well tolerated in the environment of a pH value of 3.0 and 0.1%bile salt,and had a high survival rate in artificial intestinal fluid.The strains displayed strong auto-aggregation capacity and hydrophobicity above 55%,potentially associated with their surface S-layer structures.The results of functional gene annotation revealed that the strains carried various probiotic-related functional genes,indicating robust environmental adaptability and safety.The study indicates that four Lactobacillus crispatus strains possess excellent gastrointestinal adaptability and colonization capacity,can be used as candidate strain for probiotics for poultry.
