棉籽糖对益生菌增殖作用和生长速率的影响

摘要 试验旨在通过体外试验系统评价棉籽糖作为益生元对4种益生菌增殖和生长速率的影响。采用光电比浊法测定棉籽糖对植物乳杆菌、嗜酸乳杆菌、青春双歧杆菌及婴儿双歧杆菌的体外增殖作用,并通过测定其生长过程中pH值及625 nm处的吸光度(OD_(625 nm)值)变化评价其生长速率。结果显示,培养24 h时,婴儿双歧杆菌和青春双歧杆菌达到最大增殖,培养48 h时植物乳杆菌、嗜酸乳杆菌达到最大增殖。培养48 h时,当D-果糖添加量为1.0%时,嗜酸乳杆菌的OD值最高,为1.681,植物乳杆菌、婴儿双歧杆菌和青春双歧杆菌的OD值分别为0.981、0.855和0.856;棉籽糖添加量为1.0%时,嗜酸乳杆菌的OD值最高,为1.906,植物乳杆菌、婴儿双歧杆菌和青春双歧杆菌的OD值分别为1.050、0.990和0.984。培养时间达到20 h时,植物乳杆菌达到最佳生长量;当培养时间达到48 h时,嗜酸乳杆菌达到了最佳生长量;青春双歧杆菌和婴儿双歧杆菌达到最佳生长量的时间为32 h,且4种益生菌在生长过程中不断产酸导致pH值持续下降。研究表明,棉籽糖对植物乳杆菌、嗜酸乳杆菌、青春双歧杆菌及婴儿双歧杆菌体外增殖均具有促进作用,其中对嗜酸乳杆菌的促进效果最佳。 The study aimed to systematically evaluate the effect of raffinose as a prebiotic on the proliferation and growth rate of four probiotics through in vitro experiments.Photoelectric turbidimetry was employed to measure its effects on the proliferation of Lactobacillus plantarum,Lactobacillus acidophilus,Bifidobacterium adolescentis,and Bifidobacterium infantis in vitro.Changes in pH and optical density at 625 nm(OD_(625 nm))during growth were monitored to assess its growth rate.The results showed that Bifidobacterium infantis and Bifidobacterium adolescentis reached maximum proliferation at 24 hours of cultivation,while Lactobacillus plantarum and Lactobacillus acidophilus peaked at 48 hours.At 48 hours,with 1.0%D-fructose supplementation,Lactobacillus acidophilus exhibited the highest OD value of 1.681,whereas Lactobacillus plantarum,Bifidobacterium infantis,and Bifidobacterium adolescentis showed OD values of 0.981,0.855,and 0.856,respectively.At 1.0%raffinose supplementation,Lactobacillus acidophilus achieved the highest OD value of 1.906,while Lactobacillus plantarum,Bifidobacterium infantis,and Bifidobacterium adolescentis displayed OD values of 1.050,0.990,and 0.984,respectively.Lactobacillus plantarum reached its maximum growth at 20 hours,Lactobacillus acidophilus at 48 hours,and Bifidobacterium adolescentis and Bifidobacterium infantis at 32 hours.Continuous acid production by all four probiotics led to a sustained decline in pH value during growth.The study indicates that raffinose promotes the in vitro proliferation of these probiotics,with the most significant effect observed on Lactobacillus acidophilus.
