摘要
本文主要研究甘草酸(glycyrrhizic acid,GA)纳米药物的制备及其表征。以巨大芽孢杆菌(Bacillus megaterium,简称BM)芽孢肽聚糖为载体,3-氨基苯硼酸(3-aminophenylboronic acid,APBA)为连接子,采用碳二亚胺法将APBA负载于BM芽孢肽聚糖上,获得负载前驱体,再将负载前驱体和GA混合于缓冲液中,反应制备得甘草酸纳米药物BM-APBA-GA。通过透射电镜观察BM芽孢肽聚糖的形态,傅里叶变换红外光谱和热重分析检测BM-APBA-GA的成功构建,并利用动态激光散射技术测量其粒径大小;进一步通过溶血实验验证其生物相容性,体内毒性实验验证其安全性。结果表明,以BM为载体成功构建了甘草酸纳米药物BM-APBA-GA,该纳米药物生物相容性好,并且对体内的组织器官无毒性。
This study focuses on the preparation and characterization of glycyrrhizic acid(GA)nanoparticle drug.Bacillus megaterium(BM)spore peptidoglycan was employed as a carrier,with 3-aminophenylboronic acid(APBA)as the linker.The APBA was conjugated to the BM spore peptidoglycan by the carbodiimide method to obtain the loaded precursor.Subsequently,the precursor and GA were mixed in the buffer solution to synthesize the GA nanoparticle drug,denoted as BM-APBA-GA.The morphology of BM spore peptidoglycan was observed by transmission electron microscopy(TEM),and the successful construction of BM-APBA-GA was confirmed by Fourier transform infrared spectroscopy(FTIR)and thermogravimetric analysis(TGA).The particle size was measured by dynamic laser scattering(DLS).Furthermore,the biocompatibility was assessed by hemolysis assay,and the safety was evaluated by in vivo toxicity studies.The results demonstrated that the GA nanoparticle drug BM-APBA-GA was successfully constructed with BM as a carrier.The nanoparticle drug exhibited good biocompatibility and no toxicity to the tissues and organs in vivo.
