摘要
目的:利用生物信息学方法系统探讨MPV17在肝细胞癌中的临床意义及其潜在的作用机制。方法:通过TCGA数据库下载MPV17的表达数据及肝细胞癌样本临床信息。采用Wilcoxon秩和检验比较癌组织和正常组织中MPV17的表达,采用卡方检验分析MPV17表达水平与临床病理特征的关系。采用Cox回归和Kaplan-Meier方法评估MPV17的预后价值。分析MPV17与免疫细胞浸润以及免疫检查点之间的相关性。结果:MPV17在包括肝细胞癌在内的大多数癌症中表达升高,而在少数癌症中下调,包括肾嫌色细胞癌(KICH)、胰腺癌(PAAD)、嗜铬细胞瘤和副神经瘤(PCPG)。卡方检验结果显示MPV17表达与年龄、组织学等级、AFP水平和血管侵犯密切相关。Cox回归分析显示T分期越晚和MPV17高表达是肝细胞癌患者预后的独立因素。MPV17表达与免疫细胞浸润水平以及免疫检查点密切相关。结论:MPV17在肝细胞癌中高表达,并且其高表达与肝细胞癌患者的预后不良相关。MPV17的表达与免疫细胞浸润及免疫检查点的水平密切相关,因此,MPV17可能作为肝细胞癌的诊断、预后评估以及免疫治疗的新型生物标志物。
Objective To systematically explore the clinical significance and potential mechanisms of MPV17 in hepatocellular carcinoma(LIHC)using bioinformatics.Methods The expression data of MPV17 and the clinical information of LIHC samples were downloaded from the TCGA database.The expression of MPV17 in cancerous and normal tissues was com-pared by the Wilcoxon rank-sum test,and the relationship between MPV17 and clinicopathological features was analyzed by the Chi-square test.The prognostic value of MPV17 was assessed using Cox regression and Kaplan-Meier method,and the correlation between MPV17 and immune cell infiltration,as well as immune checkpoints,was analyzed.Results MPV17 was found to be upregulated in most cancers,including hepatocellular carcinoma,while it was downregulated in a few cancers such as kidney chromophobe(KICH),pancreatic adenocarcinoma(PAAD),and pheochromocytoma and paraganglioma(PCPG).Chi-square test indicated that MPV17 was closely related to age,histological grade,AFP,and vascular invasion.Cox regression analysis revealed that advanced T-stage and high MPV17 were independent factors for poor prognosis in LIHC patients.Furthermore,MPV17 was closely correlated with immune cell infiltration and immune checkpoints.Conclusion MPV17 is highly expressed in LIHC patients,which is associated with poor prognosis.Additionally,it is also closely related to immune cell infiltration and immune checkpoints,suggesting that MPV17 could serve as a novel biomarker for diagnosis,prognosis,and immunotherapy in LIHC.
