摘要
目的探讨葡萄糖转运蛋白GLUT1/2/3/4在胃癌中的表达及临床意义,分析其与胃癌预后的相关性。方法检索公共数据集,分析GLUT1/2/3/4在胃癌肿瘤组织及癌旁组织中的表达情况。应用TCGA数据库、Kaplan-Meier Plotter在线数据平台对GLUT1/2/3/4进行受试者操作特征(ROC)曲线分析及生存分析。通过proteinatlas数据库检索GLUT1在胃癌中的定位,通过共聚焦显微镜观察GLUT4在胃癌细胞中的定位。通过STRING在线工具构建GLUT1/4蛋白互作网络图。通过GO分析及KEGG分析预测GLUT1/4的作用通路。结果GLUT1及GLUT4在胃癌中差异表达,且具有良好的诊断价值。GLUT1/2/4高表达与胃癌预后不良相关。GLUT1主要定位在胃癌胞质和胞膜。GLUT4主要定位在胃癌细胞胞质中。通路富集分析提示糖代谢和脂代谢等通路可能被激活。结论GLUT1/4可作为胃癌诊断及预后的新生物指标,在胃癌发生发展过程中的作用机制需进一步探究。
Objective To investigate the expression and clinical significance of glucose transporters GLUT1/2/3/4 in gastric cancer(GC)and analyze their correlation with prognosis.Methods Public datasets were retrieved to analyze the expression of GLUT1/2/3/4 in GC tissues and adjacent normal tissues.The TCGA database and the Kaplan-Meier Plotter online platform were used for receiver operating characteristic(ROC)curve analysis and survival analysis of GLUT1/2/3/4.The Protein Atlas database was searched to determine the subcellular localization of GLUT1 in GC,while confocal microscopy was employed to observe the localization of GLUT4 in GC cells.The STRING online tool was utilized to construct a protein-protein interaction(PPI)network of GLUT1/4.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses were performed to predict the functional pathways of GLUT1/4.Results GLUT1 and GLUT4 were differentially expressed in GC and demonstrated good diagnostic value.High expression of GLUT1/2/4 was associated with poor prognosis in GC.GLUT1 was primarily localized in the cytoplasm and cell membrane of GC cells,whereas GLUT4 was mainly found in the cytoplasm.Pathway enrichment analysis suggested the potential activation of glucose and lipid metabolism-related pathways.Conclusion GLUT1/4 may serve as novel biomarkers for the diagnosis and prognosis of GC,but their mechanistic roles in GC progression require further investigation.
