摘要
为探明养殖条件下成年(4+龄)大鳍鳠精巢发育规律及血清性类固醇激素(睾酮、雌二醇和11-酮基睾酮)含量的周年变化特征,通过组织切片技术和酶联免疫法,对养殖条件下成年大鳍鳠精巢发育的形态及组织学特点和血清中性类固醇激素含量的周年变化进行分析。实验结果表明,养殖条件下成年大鳍鳠精巢扁平,有着明显的指状分枝,其周年变化经历如下过程:4月为Ⅳ期精巢(精子开始出现期),5月发育至Ⅴ期精巢(完全成熟期),7月绝大部分发育至Ⅴ期精巢,8~12月均有Ⅴ期、Ⅵ期(退化吸收期)精巢存在。翌年3月发育至Ⅲ期(精母细胞成熟期)。养殖条件下成年大鳍鳠GSI、HSI、CF的周年变化范围分别为0.06%~0.38%、0.77%~2.15%、0.61%~0.88%,其血清睾酮水平的变化同精巢发育基本一致,在精子排放过程中达到最大值(8.50μg/L);11-酮基睾酮在精子排放结束后达到最大值(0.22μg/L),雌二醇在睾酮峰值之后达到峰值(43.74 ng/L)。首次对全人工养殖条件下的成年大鳍鳠雄鱼性腺发育进行了观察和研究,相关数据可为掌握大鳍鳠的繁殖规律和优化人工繁育技术提供理论参考。
The annual maturation cycle of testis and variation in sex steroids hormone level in adult Mystus macropterus were investigate by histological sectioning and enzyme immunoassay.The experimental results show that the spermathecae of adult M.macropterus were flattened under culture conditions,had obvious finger-like branches,and their annual changes went through the following process:Ⅳtestis(sperms begin to appear)in April,Ⅴtestis(complete maturation of sperm)in May,the vast majority of them developed toⅤtestis in July.ⅤandⅥstages(degradation of sperm)of testis were present from August to December.In March of the following year,it develops to stageⅢ(spermatocytes mature).In captivity,the annual variation of GSI ranged 0.06%~0.38%,HSI ranged 0.77%~2.15%,and CF ranged 0.61%~0.88%.Serum testosterone levels were synchronized with testis development,reaching a maximum value(8.50μg/L)during sperm emission.The 11-ketotestosterone peaked(0.22μg/L)after sperm emission cessation,while estradiol attained its maximum(43.74 ng/L)following the testosterone peak.This study is the first to observe and study the gonadal development of adult M.macropterus in artificial breeding conditions.The relevant data can provide theoretical a reference for mastering the reproductive rules of M.macropterus and optimizing the artificial breeding technology.
