基于非靶向代谢组学筛选‘泽州芹菜’抗虫物质

Screening of Insect Resistant Substances of ‘Zezhou Celery’ Based on Non-targeted Metabolomics

摘要 以‘泽州芹菜’和‘多利王芹菜’为试材,采用基于液质联用(LC-MS/MS)非靶向代谢组学方法,研究了‘多利王芹菜’幼苗期(CSY)、‘泽州芹菜’幼苗期(ZSY)和开花期(ZS)叶片之间的差异代谢物,以期明确‘泽州芹菜’抗虫相关的关键物质。结果表明:ZSY_vs_CSY组共筛选出31个差异代谢物,ZS_vs_ZSY组共筛选出141个差异代谢物。这些差异代谢物包括脂质和类脂分子、苯丙类和聚酮类、有机酸及其衍生物、有机氧化合物和有机杂环化合物等。其中脂质和类脂分子中萜类、苯丙类和聚酮类中黄酮类是起主要抗虫作用的物质,进一步对萜类和黄酮类研究发现,与CSY相比,ZSY中2,6-二叔丁基苯醌、石竹稀氧化物含量显著增加,茶黄素含量显著下降;与ZSY相比,ZS中芳樟醇含量显著增加,芹菜素、柚皮素、2,6-二叔丁基苯醌、β-石竹烯、三尖杉宁碱和槲皮素含量显著下降。这9种代谢物对提高‘泽州芹菜’抗虫性均具有不同程度的作用,这些物质的发现为开发新型绿色农药提供参考依据,有助于减少化学农药的使用,推动农业的可持续发展。 Taking‘Zezhou Celery’and‘Dory King Celery’as the test materials,the differential metabolites among the leaves of‘Duoliwang Celery’at the seedling stage(CSY),‘Zezhou Celery’at the seedling stage(ZSY),and flowering stage(ZS)were studied by using the non-targeted metabolomics method based on liquid chromatography-mass spectrometry(LC-MS/MS),in order to clarify the key substances related to insect resistance of‘Zezhou Celery’.The results showed that 31 different metabolites were screened in the ZSY_vs_CSY group and 141 different metabolites were screened in the ZS_vs_ZSY group.These differential metabolites included lipid and lipid molecules,phenylpropyl and polyketones,organic acids and their derivatives,organic oxygen compounds and organic heterocyclic compounds.Among them,terpenoids in lipids and lipids,and flavonoids in phenylpropyl and polyketones were the main substances that play anti-insect roles,further studies on terpenoids and flavonoids showed that compared with CSY,the contents of 2,6-Di-tert-butyl-1,4-benzoquinone and(-)-Caryophyllene oxide in ZSY were significantly increased,while theaflavin content was significantly decreased.Compared with ZSY,the contents of linalool in ZS increased significantly,while the contents of apigenin,naringin,2,6-Di-tert-butyl-1,4-benzoquinone,β-caryophyllene,cephalomannine and quercetin decreased significantly.These 9 kinds of metabolites have different degrees of effect on improving the pest resistance of‘Zezhou Celery’.The discovery of these substances provided a reference for the development of new green pesticides,helped to reduce the use of chemical pesticides and promote the sustainable development of agriculture.
