摘要
本研究以桉叶油醇、α-松油醇为研究对象,探究化合物单体及复配对赤拟谷盗(Tribolium castaneum)的杀虫作用机制。结果显示,桉叶油醇、α-松油醇对赤拟谷盗具有触杀作用,且二者4∶6复配时杀虫效果增强,LD 50为20.30μg/头;结合分子对接技术对赤拟谷盗解毒酶进行筛选,发现桉叶油醇、α-松油醇及复配对代谢型谷氨酸受体样蛋白(TcMGRP)结合较为稳定,结合能分别为-6.53、-6.79、-11.49 kJ/moL;此外,实时荧光定量PCR(RT-PCR)结果显示,随着桉叶油醇、α-松油醇单体及复配(4∶6)处理时间的延长,TcMGRP的基因表达量逐渐下降,呈时间依赖性,而且,在6、12 h时,桉叶油醇、α-松油醇4∶6复配作用下赤拟谷盗TcMGRP基因表达量显著高于单体化合物的作用(P<0.05)。桉叶油醇、α-松油醇单体及复配(4∶6)通过抑制赤拟谷盗TcMGRP基因的表达发挥杀虫作用。
In this study,eucalyptol andα-terpineol were used as research objects to explore the insecticidal mechanism of single compound and their complex.The results indicated that eucalyptol andα-terpineol exhibited contact effect on Tribolium castaneum,and the insecticidal effect was enhanced when the these two were mixed in a 4∶6 ratio,with the LD 50 value of 20.30μg/adult.Combined with molecular docking technology,detoxification enzymes were screened for Tribolium castaneum,and it was found that eucalyptol,α-terpineol and the complex could be bound stably with metabotropic glutamate receptor-like protein(TcMGRP),with affinity scores of-6.56,-6.79 kJ/moL and-11.49 kJ/moL,respectively.In addition,the results of the real-time quantitative PCR(RT-PCR)revealed that the gene expression of TcMGRP decreased in a time-dependent manner as the treatment time of eucalyptol,α-terpineol and the complex(4∶6)increased.Besides,the gene expression of TcMGRP under the complex of eucalyptol andα-terpineol with a 4∶6 ratio treatment was significantly higher than that of the single compound at 6 h and 12 h(P<0.05).Studies have shown that eucalyptol,α-terpineol and their complex exhibited insecticidal effects by inhibiting the expression of TcMGRP.
