摘要
本文建立了超高效液相色谱-三重四极杆串联质谱对烟草中4种原花青素进行定性定量分析的方法。烟草样品经冷冻干燥研磨后,以80%的乙腈-水溶液提取,Carbon分散吸附剂净化后,采用超高效液相色谱-三重四极杆串联质谱法,以多重反应监测负离子模式检测,内标法定量。结果表明:儿茶素与原花青素B_(2)在1.0~500μg/L、表儿茶素在5.0~500μg/L、原花青素B_(1)在2.0~500μg/L浓度范围内线性良好(r^(2)>0.999),检出限为0.25~1.0μg/L。3个浓度添加水平下,4种原花青素的回收率为94.5%~111.2%,相对标准偏差为0.78%~9.88%。该方法简单快速、灵敏准确,适合烟草中4种原花青素的快速测定。
An analytical method based on ultra-high performance liquid chromatography-triple quadrupole tandem mass spectrometry(UPLC-MS/MS)was proposed for the simultaneous determination of four procyanidins in tobacco.The tobacco sample was ground by freeze-drying,extracted with acetonitrile-water 80%(V/V)solution and cleaned up by Carbon solid-phase dispersion.The extracts were subjected to UPLC-MS/MS analysis with negative ion modes and multiple reaction monitoring(MRM)mode,and four target procyanidins were quantified by using internal standard method.The proposed method exhibited correlation coefficients(r^(2))of more than 0.999 in the range of 1.0-500 ng/mL for catechin and procyanidin B_(2),5.0-500 ng/mL for epicatechin,and 2.0-500 ng/mL for procyanidin B_(1),with the limits of detection(LOD)of 0.25-1.0μg/L.The recoveries of target procyanidins in tobacco sample extracts were in the range of 94.5%-111.2%,with the relative standard deviations of 0.78%-9.88%.The method is simple,rapid,sensitive,accurate,and suitable for the quantification of four procyanidins in tobacco.
