黑木耳栽培菌株遗传多样性研究

摘要 以27株黑木耳栽培菌株为试材,通过拮抗试验及ITS、RAPD和ISSR聚类分析,探究供试菌株间的亲缘关系。结果表明,27株黑木耳菌株两两之间均有拮抗反应,主要分为隔离型、隆起型和隆起型且有色素沉积;RAPD和ISSR引物扩增条带多态性比率分别为95.7%和100%,相似系数大于0.95的菌株分别占37.0%和44.4%;ITS、RAPD和ISSR聚类分析均将27株黑木耳菌株分为四大类,H28和H31与其他菌株亲缘关系较远,其余菌株间遗传差异较小,遗传多样性欠佳。 Using 27 strains of cultivated Auricularia heimuer as materials,the genetic relationship of 27 Auricularia heimuer was wxplored by antagonistic test and cluster analysis of ITS,RAPD and ISSR.The results showed that antagonistic reaction existed among all strains,mainly showed isolate type,uplift type and uplift type with pigmentation.The RAPD and ISSR primers amplified bands polymorphism ratio were 95.7%and 100%,the strains with similarity coefficient greater than 0.95 accounted for 37.0%and 44.4%.ITS,RAPD and ISSR clustering analysis showed that the 27 strains could be divided into four categories,the genetic relationship between H28,H31 and other strains was far.There were little genetic difference and poor genetic diversity among the other strains.
