摘要
介绍了鸡蛋粉中8种氟喹诺酮基体标准物质的研制过程。采用自主加标的方式,对市售新鲜鸡蛋进行初检,选用未检出目标物的鸡蛋为直接原料,采用分离、混合、加标、冻干、过筛等技术手段引入氧氟沙星、诺氟沙星、洛美沙星等8种氟喹诺酮并使其固定在鸡蛋粉末中进行制备,通过液相色谱-串联质谱法和超高效液相色谱法对加标原料进行了外标法定值,对其特性量值进行均匀性及稳定性检验,并系统分析与评估了鸡蛋粉中8种氟喹诺酮类基体标准物质研制过程中所引入的不确定度。研制的鸡蛋粉中氧氟沙星、诺氟沙星、培氟沙星、洛美沙星、环丙沙星、达氟沙星、恩诺沙星、沙拉沙星基体标准物质定值结果分别为(86±8.8)、(88±8.7)、(83±8.6)、(92±8.5)、(93±9.2)、(94±8.5)、(93±8.7)、(95±9.0)μg/kg,研制流程与结果符合标准物质研制定值要求。该标准物质可用于实验室内部质量控制的实施,包括但不限于分析方法验证、检测过程的质量保证。
The development process of eight fluoroquinolone matrix reference materials in egg powder was introduced.Using the method of independent labeling,the initial inspection of fresh eggs on the market was carried out.Eggs that did not detect the target substance were selected as direct raw materials.Eight fluoroquinolones,including ofloxacin,norfloxacin,and lomefloxacin,were introduced and fixed in egg powder through separation,mixing,labeling,freeze-drying,screening,and other technical means to prepare the reference materials.The external standard values of the labeled raw materials were determined by liquid chromatography tandem mass spectrometry and ultra-high performance liquid chromatography,and the uniformity and stability of their characteristic values were tested.The uncertainty introduced in the development process of eight fluoroquinolone matrix standard substances in egg powder was systematically analyzed and evaluated.The determination values of ofloxacin,norfloxacin,levofloxacin,lomefloxacin,ciprofloxacin,norfloxacin,enrofloxacin,and sarafloxacin matrix standard substances in the developed egg powder are(86±8.8),(88±8.7),(83±8.6),(92±8.5),(93±9.2),(94±8.5),(93±8.7),and(95±9.0)μg/kg,respectively.The development process and results meet the requirements of standard substance determination.This standard substance can be used for internal quality control in the laboratory,including but not limited to quality assurance of analytical method validation and testing processes.
