山羊源化脓隐秘杆菌的分离鉴定及生物学特性分析

摘要 为确定导致山羊肝化脓性病变的病原菌,无菌采集化脓性肝炎病灶部位样本,进行细菌分离培养,通过细菌形态学观察和16S rRNA基因测序鉴定细菌种类,并对分离菌株进行毒力基因检测、药敏试验以及对小鼠的致病性试验分析。结果显示:从化脓性肝组织中分离鉴定出1株化脓隐秘杆菌,该分离株携带nanH、nanP、fimA、fimC、plo等毒力基因;该分离株对阿米卡星、妥布霉素、氯霉素、氧氟沙星、麦迪霉素、克拉霉素、万古霉素、头孢哌酮和头孢西丁高度敏感,对复方新诺明、多黏菌素B和大部分β-内酰胺类抗生素存在耐药性;该分离株可使试验小鼠肝发生化脓性病变。综上所述,成功地鉴定出化脓隐秘杆菌是导致山羊化脓性肝炎的病原,为研究化脓性杆菌的致病机制提供了参考,也为临床治疗化脓隐秘杆菌感染动物提供了科学依据。 To identify the bacterum responsible for suppurative hepatic lesion in goat,samples were collected from purulent hepatitis tissue for isolation,and the pathogen was identified using bacterial culture,morphology observation and 16S rRNA gene sequencing.The virulence gene profiling and antimicrobial susceptibility of the isolated bacterium were tested.In addition,experimental pathogenicity was also evaluated in a mouse model.The results showed that a Trueperella pyogenes strain was isolated and identified from the suppurative goat liver.This isolate carried several virulence genes,including nanH,nanP,fimA,fimC and plo,and was highly sensitive to amikacin,tobramycin,chloramphenicol,ofloxacin,medemycin,clavicycin,vancomycin,cefoperazone and cefoxitin.However,the isolate was resistant to cotrimoxazole,polymyxin B and mostβ-lactam antibiotics.Moreover,the isolate exhibited pathogenicity to mice which showed suppurative lesions in the liver when inoculated with the bacterium.In summary,this study demonstrated that T.pyogenes was the pathogen responsible for purulent hepatitis in goats.This study provided clues to further understanding of the pathogenic mechanism of T.pyogenes and also provided a basis for clinical treatment of animals infected with this bacterium.
