摘要
目的探究黄芪润肠方对气阴两虚型便秘小鼠便秘的疗效及作用机制。方法选取60只雄性C57BL/6J小鼠,随机分为对照组、便秘组、黄芪润肠方低、中、高剂量组及阳性对照组,每组10只。除对照组外,其他5组均通过饮食不节+热性药灌胃+负重游泳法建立气阴两虚型便秘模型。造模后,黄芪润肠方低、中、高剂量组分别给予0.0915、0.183、0.366 g/(kg·d)黄芪润肠方灌胃,阳性对照组给予6.2 mL/(kg·d)乳果糖口服溶液灌胃,对照组和便秘组给予等容积0.9%氯化钠注射液灌胃,连续灌胃14 d。比较各组排便参数(排出首粒黑便的时间、粪便含水量和粪便总数)、胃肠蠕动相关生化指标[胃动素(MTL)、胃泌素(Gas)、内皮素1(ET-1)、P物质(SP)、血管活性肠肽(VIP)]、肝肾功能指标[天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、血白蛋白(ALB)和血肌酐(SCr)]、小肠组织病理变化及紧密连接相关蛋白[紧密连接蛋白1(Claudin-1)、闭合蛋白(Occludin)和闭锁小带蛋白-1(ZO-1)]表达。结果与便秘组比较,黄芪润肠方中、高剂量组和阳性对照组排出首粒黑便时间显著降低(P<0.05),粪便总数和粪便含水量增加(P<0.05);血清MTL、Gas、SP和ET-1水平升高(P<0.05),VIP水平降低(P<0.05),小肠组织内结构损伤程度显著改善,小肠组织中Claudin-1、Occludin和ZO-1相对表达量显著升高。各组小鼠血清AST、ALT、ALB和SCr水平比较,差异无统计学意义(P>0.05)。结论黄芪润肠方可通过调控紧密连接相关蛋白表达、修复小肠黏膜与改善胃肠功能有效减轻气阴两虚型便秘小鼠症状。
Objective To explore the effect and mechanism of Huangqi Runchang prescription on constipation based on the mouse model of Qi-Yin deficiency.Methods Male C57BL/6J mice were divided into the control group,the constipation group,the Huangqi Runchang prescription low-,medium-and high-dose groups[0.0915,0.183,0.366 g/(kg·d)]and positive control group[6.2 mL/(kg·d)lactulose],with 10 mice in each group.The constipation model of Qi and Yin deficiency was established by a combination of improper diet+hot drug administration+weight-bearing swimming.After corresponding drug intervention for 14 days,the defecation parameters(time to first black stool,fecal water content and total fecal matter)were compared among the groups.The following parameters were analyzed:gastrointestinal motility biochemical indexes[motilin,gastrin,endothelin,Substance P,and vasoactive intestinal peptide];liver and kidney function indexes[aspartate aminotransferase,alanine aminotransferase,albumin and serum creatinine];small intestinal pathological changes;and expression of tight-junction related proteins.Results Compared with the constipation group,the time of first black stool excretion in the Huangqi Runchang prescription medium-and high-dose groups and the positive control group was significantly decreased,while total feces output and fecal water content were increased(P<0.05).Serum levels of motilin,gastrin,Substance P,and endothelin-1 increased(P<0.05),whereas vasoactive intestinal peptide levels decreased significantly(P<0.05).There were no significant differences in the serum levels of aspartate aminotransferase,alanine aminotransferase,albumin,or creatinine among all groups.The degree of structural damage in small intestinal tissue was significantly improved.The relative expression levels of occludin,claudin-1 and zonula occludens-1 in the small intestine increased.Conclusion Huangqi Runchang prescription can effectively relieve Qi-Yin deficiency-induced constipation symptoms in mice by regulating tight-junction-relat-ed protein expression,repairing the small intestinal mucosa,and improving gastrointestinal function.
