基于代谢组学探讨麻杏石甘汤调节肺炎链球菌肺炎的潜在机制

Exploration of the potantial mechanisms of Maxing Shigan Decoction on regulating Streptococcus pneumoniae pneumonia based on metabolomics methods

摘要 目的 使用代谢组学方法探讨麻杏石甘汤靶向PD-1/PD-L1调节肺炎链球菌肺炎的机制。方法 使用肺炎链球菌D39滴鼻造模,HE染色检测麻杏石甘汤对小鼠肺部炎症的治疗作用,基于液质联用(LC-MS)技术进行非靶向代谢组学分析,差异代谢物使用VIP、FC和p-value进行筛选,最后使用KEGG分析并筛选主要靶通路和代谢物。结果 PD-1/PD-L1抗体加重肺部炎症,麻杏石甘汤可减少炎症反应。麻杏石甘汤或可通过嘧啶代谢、FoxO信号通路、花生四烯酸代谢和脂肪酸生物合成调节PD-1/PD-L1通路和肺炎链球菌肺炎的交互作用。结论 麻杏石甘汤可调节PD-1/PD-L1通路治疗肺炎链球菌肺炎。 Objective To explore the mechanisms of Maxing Shigan Decoction targeting PD-1/PD-L1 on regulating Streptococcus pneumoniae pneumonia using metabolomics methods.Methods Streptococcus pneumoniae D39 was used by nasal drip to create a model,and HE staining was used to detect the therapeutic effect of Maxing Shigan Decoction in the treatment of pulmonary inflammation in mice.Non-targeted metabolomics analysis was performed based on the liquid chromatography-mass spectrometry(LC-MS)technology,and differential metabolites were screened using VIP,FC,and p-value.Finally,KEGG analysis was conducted to screen the main target pathways and metabolites.Results PD-1/PD-L1 antibodies aggravated lung inflammation,while Maxing Shigan Decoction reduced inflammatory responses.Maxing Shigan Decoction possibly regulated the interaction between the PD-1/L1 pathway and Streptococcus pneumoniae pneumonia through pyrimidine metabolism,FoxO signaling pathway,arachidonic acid metabolism and fatty acid biosynthesis.Conclusion Maxing Shigan Decoction can regulate the PD-1/PD-L1 pathway to treat Streptococcus pneumoniae pneumonia.
