台湾乳白蚁工蚁和兵蚁的转录组测序比较及表达分析

Comparative transcriptome sequencing and expression analysis ofworkers and soldiers of Coptotermes formosanus

摘要 目的 了解台湾乳白蚁Coptotermes formosanus Shiraki工蚁和兵蚁的转录组特征,为阐明工蚁和兵蚁品级分化的分子机制提供理论依据。方法 对工蚁和兵蚁的头部组织进行转录组测序,对比两者之间的差异表达基因(DEGs),并进行京都基因与基因组百科全书(KEGG)数据库信号通路和基因本体(GO)数据库功能富集分析。结果 台湾乳白蚁转录组测序数据为39.13 Gb,经质控过滤后得到262 172 274条clean reads, GC含量为38.69%~41.27%。工蚁和兵蚁有996个DEGs。GO功能注释结果显示,涉及生物过程的DEGs最多;KEGG信号通路富集结果显示,工蚁和兵蚁的DEGs显著(Padjust<0.05)富集在单磷酸腺苷激活的蛋白激酶(AMPK)信号通路。结论 工蚁和兵蚁在基因上的差异对应了它们各自的社会分工,差异基因的上调基因与免疫防御有关,下调基因与能量代谢和蛋白合成相关。 Objective To understand the transcriptome characteristics of workers and soldiers of Coptotermes formosanus Shiraki,so as to to provide a theoretical reference for clarifying the molecular mechanism of caste differentiation between workers and soldiers.Methods Transcriptome sequencing was performed on the head tissues of workers and soldiers.Differentially expressed genes(DEGs)of the two castes were compared,and Gene Ontology(GO)functional annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)signal pathway enrichment analysis were conducted to analyze DEGs.Results Transcriptome sequencing data of C.formosanus was 39.13 Gb,and 262172274 clean reads were obtained after quality control filtering.The GC content ranged from 38.69%to 41.27%.There were 996 DEGs between workers and soldiers.GO functional annotation results revealed that the majority of the DEGs were associated with biological processes.The results of KEGG signal pathway enrichment analysis showed that DEGs of workers and soldiers were significantly enriched(P adjust<0.05)in the AMPK signaling pathway.Conclusion The genetic differences between workers and soldiers correspond to their respective social divisions of labor.The up-regulated genes were related to immune defense,and the down-regulated genes were related to energy metabolism and protein synthesis.
