摘要
为建立一种以单壁碳纳米管(SWCNTs)作为吸附剂,结合高效液相色谱仪同时测定鸡肉中喹诺酮类和磺胺类药物残留的快速基质萃取方法,精确称取鸡肉样品2.0g,用2%乙酸-乙腈提取,以SWCNTs净化,5%氨水-甲醇溶液洗脱待测药物,经衍生化处理后利用高效液相色谱仪-荧光检测器检测。结果显示:22种药物在0.5~400μg/L范围内线性关系良好,相关系数(R^(2))大于0.999;22种药物的检测限为0.5~2.5μg/kg;空白添加试验中,药物的平均回收率为66.6%~104.4%,批内变异系数为2.9%~14.6%,批间变异系数为4.5%~18.1%。结果表明,本研究建立的方法操作简便、耗时短,精密度与准确度良好,可满足鸡肉中喹诺酮类和磺胺类药物残留的检测要求.
In order to simultaneously determine quinolone and sulfonamide residues in chicken by a rapid matrix extraction assay that was developed taking single-walled carbon nanotubes(SWCNTs)as adsorbents,and in combination with high-performance liquid chromatography(HPLC).2.0 g of chicken sample was accurately weighed,extracted with 2%acetic acid-acetonitrile solution,and then purified using SWCNTs.The target analytes were eluted with 5%ammonia-methanol solution,and detected by HPLC with a fluorescence detector(HPLC-FLD)after derivatization.The results showed that,for 22 drugs,good linearity was observed within the range of 0.5 to 400μg/L,with correlation coefficients(R^(2))>0.999;the limits of detection(LODs)ranged within 0.5 to 2.5 ug/kg:and an average recovery rate ranging from 66.6%to 104.4%was found in spiked recovery test,with intra-assay coefficient of variation of 2.9%to 14.6%and inter-assay coefficient of variation of 4.5%to 18.1%.In conclusion,the method established in the study,benefiting from its user-friendly control,time-efficiency,good precision and accuracy,could be used to detect quinolone and sulfonamide residues in chicken.
