免疫细胞与尘肺病之间的因果关系:一项孟德尔随机化研究

摘要 目的:通过孟德尔随机化(MR)方法找出免疫细胞与尘肺病之间潜在的因果联系。方法:基于已公开的全基因组关联研究(GWAS),采用两样本MR分析731个免疫细胞标签与尘肺病的因果关系,另外采用5种MR分析方法及综合敏感性分析评价结果的可靠性。结果:在α=0.05条件下,CD45RA+CD4^(+)T细胞上的CD127(P=0.007)和CD4^(+)T细胞上的CD4(P=0.029)两种免疫细胞表型是尘肺病的危险因素,但未发现对尘肺病有保护作用的免疫细胞表型。调整显著性为0.20时,发现2种免疫细胞表型是尘肺病的危险因素,即人类白细胞抗原(HLA)DR+CD4^(+)T细胞表面的CD4(P=0.189)和初始CD4^(+)T细胞表面的CD4(P=0.152);IgD+CD24+B细胞上的IgD(P=0.133)和CD33+HLA DR+CD14dim(P=0.148)两种免疫细胞表型是尘肺病的保护因素。结论:从遗传学角度证明免疫细胞与尘肺病之间的密切联系,提示使用相关药物调节特异性免疫细胞功能可能有益于尘肺病的治疗。
