摘要
目的探讨原发性干燥综合征(primary Sjögren syndrome,PSS)患者外周血T细胞受体(T cell receptor,TCR)ζ链及Zeta链相关蛋白激酶70(Zeta-chain-associated protein kinase 70,ZAP70)的表达水平。方法选取2024年1月至6月于九江市第一人民医院就诊的PSS患者36例纳入观察组,同期健康体检者30名纳入对照组。采用实时荧光定量聚合酶链反应检测外周血单个核细胞中TCRζ链及ZAP70的表达水平,流式细胞术检测外周血T细胞亚群。采用Pearson相关分析TCRζ链、ZAP70与其他检测指标的相关性。结果观察组患者的TCRζ链、ZAP70相对表达量均显著低于对照组(P<0.05),CD8^(+)、白细胞介素-6(interleukin-6,IL-6)均显著高于对照组(P<0.05)。Pearson相关分析显示:TCRζ链与CD4^(+)呈正相关,与CD8^(+)、IL-6呈负相关(P<0.05);ZAP70与CD8^(+)、IL-6呈负相关(P<0.05)。结论PSS患者的TCRζ链及ZAP70表达均下调,可能通过T细胞信号传导异常加剧PSS的免疫紊乱。
Objective To investigate the expression levels of T cell receptor(TCR)ζchain and Zeta-chain-associated protein kinase 70(ZAP70)in peripheral blood of patients with primary Sjögren syndrome(PSS).Methods Thirty-six patients with PSS who were treated at Jiujiang NO.1 People’s Hospital from January to June 2024 were enrolled in observation group,and 30 healthy subjects during the same period were enrolled in control group.Real-time fluorescent quantitative polymerase chain reaction was used to detect the expression levels of TCRζchain and ZAP70 in peripheral blood mononuclear cells,and flow cytometry was used to detect peripheral blood T cell subsets.Pearson correlation was used to analyze the correlation between TCRζchain,ZAP70 and other detection indicators.Results The relative expression levels of TCRζchain and ZAP70 in observation group were significantly lower than those in control group(P<0.05),while CD8^(+)and interleukin-6(IL-6)were significantly higher than those in control group(P<0.05).Pearson correlation analysis showed that TCRζchain was positively correlated with CD4^(+),and negatively correlated with CD8^(+)and IL-6(P<0.05).ZAP70 was negatively correlated with CD8^(+)and IL-6(P<0.05).Conclusion The expressions of TCRζchain and ZAP70 are down-regulated in PSS patients,which may exacerbate the immune disorder of PSS through abnormal T cell signal transduction.
