摘要
血管细胞黏附分子-1(VCAM-1)属于免疫球蛋白超家族中的细胞黏附分子的成员,包含一个具有6或7个胞外IgG样结构域、一个胞质结构域和一个跨膜结构域,其主要配体为整合素α4β1(VLA-4)。TNFα等因子会通过MAPK、NF-κB等信号通路促进VCAM-1的表达,VCAM-1与配体结合激活下游信号分子,从而促进白细胞对血管内皮的黏附和迁移,与疾病的发展和预后密切相关。研究表明VCAM-1在血栓性疾病中高表达,如动脉粥样硬化、静脉血栓栓塞等。VCAM-1在炎症性疾病如慢性炎症性疾病、脓毒症等的诊断和治疗中同样发挥重要作用。本综述涵盖VCAM-1在调控机体生理和病理状态中的作用,并阐明VCAM-1在疾病诊断与作为新治疗靶点中的巨大潜力。
Vascular cell adhesion molecule-1(VCAM-1)is a member of the immunoglobulin superfamily,containing an extracellular domain with 6 or 7 immunoglobulin(Ig)-like domains,a cytoplasmic domain and a transmembrane domain.Its primary ligand is integrinα4β1(VLA-4).TNF-αand other factors induce the expression of VCAM-1 via MAPK,NF-κB and other signaling pathways.VCAM-1 binds to ligands and activates downstream signaling molecules,thereby enhancing the adhesion and migration of leukocytes to vascular endothelium,and is closely associated with the development and prognosis of the disease.Evidence indicates that the expression of VCAM-1 is increased in thrombotic diseases,such as atherosclerosis and venous thromboembolism.VCAM-1 plays a crucial role in the diagnosis and treatment of inflammatory diseases such as chronic inflammation and sepsis.This review summarizes the role of VCAM-1 in regulating physiological and pathological conditions in vivo,and highlights the therapeutic potential of VCAM-1 in diagnosing diseases and becoming a new therapeutic target.
