摘要
以抗旱高淀粉加工型马铃薯新品种蒙乌薯6号脱毒苗为试验材料,研究不同培养器皿对脱毒苗长势的影响,筛选益培隆抑菌剂防治细菌污染有效使用浓度及品种容器薯适宜的诱导方法。结果表明,全自动配套塑料方盒处理下的脱毒苗植株粗壮,光合效率高,长势强,能够提高马铃薯脱毒苗扩繁倍数,优于玻璃瓶培养;蒙乌薯6号细菌防治最适宜的益培隆使用浓度(w,后同)为0.20%;容器薯快速诱导培养方法为固体(壮苗阶段:MS+30 g·L^(-1)蔗糖+6 g·L^(-1)卡拉胶)+液体(容器薯诱导阶段:MS+80 g·L^(-1)蔗糖+500 mg·L^(-1)矮壮素+5 mg·L^(-1)6-苄氨基嘌呤)。研究结果不仅为蒙乌薯6号产业化发展奠定了技术基础,而且为同类型马铃薯品种研究提供了参考依据。
Using virus-free seedlings of the new drought-resistant and high-starch processing potato variety Mengwushu No.6 as experimental material,this study investigated the effects of different culture vessels on the growth of variety virus-free seedlings,screened the effective concentration of Yipeilong bacteriostatic agent for bacterial contamination control,and explored the suitable induction method for container potato of this variety.The results showed that the virus-free seedlings in the fully automatic plastic square box were more robust,with improved photosynthetic efficiency and stronger growth potential.This treatment could increase the multiplication coefficient of potato virus-free seedlings,which was superior to the glass bottle culture.The most suitable concentration of Yipeilong for preventing bacterial contamination in Mengwushu No.6 was 0.20%.The rapid induction culture method for in vitro potato tubers is as follows:Solid medium(seedling-strengthening stage:MS+30 g·L^(-1) sucrose+6 g·L^(-1) carrageenan)+liquid medium(container potato induction stage:MS+80 g·L^(-1) sucrose+500 mg·L^(-1) chlormequat chloride+5 mg·L^(-1)6-benzylaminopurine).The results of this study not only lay a technical foundation for the industrialization development of Mengwushu No.6,but also provide a reference for research on similar potato variety.
