摘要
为探究斑马鱼亲代氟暴露对子代(F1)的神经发育毒性,采用系列浓度的氟化钠溶液对亲本斑马鱼进行暴露实验,90 d后交配产生子代,通过体视显微镜观察子代胚胎及幼鱼的发育和行为学变化,结合组织学分析脑结构损伤,利用酶联免疫吸附法测定子代超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)、丙二醛(MDA)及神经递质多巴胺含量,此外,利用qPCR技术检测多巴胺通路相关基因的表达水平.结果表明,亲代氟暴露可诱导F1代发育毒性,使F1代的行为学和抗氧化能力受到负面影响,且随着亲本氟暴露浓度的增大,F1代脑组织病变、多巴胺含量、多巴胺通路相关基因bdnf、th的表达随亲代暴露浓度升高而下调.这些发现证实了亲代氟暴露对子代神经系统的显著毒性,为评估氟的传代健康风险提供了重要依据.
This study aimed to investigate the neurotoxic effects of fluoride exposure in zebrafish parents on F1 generation embryos/larvae.Parent zebrafish were exposed to a series of concentrations of fluoride solution for 90 days prior to generate offspring.The impacts of parental fluoride exposure on development and behavior of F1 generation embryos/larvae were observed and analyzed using a stereomicroscope.The histopathological changes in brain tissues of F1 were analyzed with histological methods.The levels of superoxide dismutase(SOD),glutathione(GSH),malondialdehyde(MDA)and dopamine content in F1 generation were quantified uing Enzyme linked immunosorbent assay(ELISA).The expression levels of dopamine related genes were assessed by real-time fluorescence quantitative PCR.The results showed that compared with the control group,parental fluoride exposure induced developmental toxicity in F1 offspring,adversely impairing their behavior and antioxidant capacity.Furthermore,as the fluoride exposure concentration in parents increased,the histopathological changes in brain tissues,dopamine levels,and the expression levels of genes such as bdnf,th,zc4h2,drd2b,drd3,and drd4b in the F1 generation showed downward trends.These findings demonstrate that parental fluoride exposure exerts substantial neurotoxic effects on F1 embryos/larvae,which provides a theoretical basis for further investigation into the toxic effects of fluoride and the assessment of its potential health risks.
