野生和栽培红花龙胆药材品质的比较研究

摘要 目的:在不同层面评价野生和栽培红花龙胆药材的品质特征,为红花龙胆的科学种植研究以及作为野生资源的替代品提供数据支持和理论依据。方法:以野生和栽培红花龙胆为对象,分别对其药材鲜品和干品的性状及显微特征进行鉴别。对芒果苷含量以及水分、总灰分及浸出物含量等药材品质指标进行测定。通过建立HPLC指纹图谱同步评价整体化学成分的相似度。结果:野生和栽培红花龙胆药材的株高、分枝数、叶长、叶宽等性状特征存在显著差异,野生药材的株高为46.80~80.93 cm,是栽培药材的1.3倍;野生药材的节间长和叶面积分别为栽培药材的1.24倍和1.17倍;但栽培药材的二级分枝数为野生药材的1.59倍。野生和栽培红花龙胆的显微特征无显著差异。以芒果苷为指标成分,栽培红花龙胆的芒果苷含量为36.76~50.70 mg/g,野生红花龙胆的芒果苷含量为38.28~51.62 mg/g,均符合2020年版中国药典的要求。HPLC指纹图谱中共确定了18个共有峰,相似度均在0.900以上。结论:野生和栽培红花龙胆除了株高和分枝数外,其余各项质量指标无显著差异,表明野生和栽培红花龙胆药材品质相似,今后应开展药效学方面的比较研究,以保障临床用药的安全有效。 Objective:To evaluate the quality characteristics of wild and cultivated Gentiana rhodantha at different levels,so as to provide data support and theoretical basis for scientific planting research of Gentiana rhodantha and as a substitute for wild resources.Methods:The wild and cultivated Gentiana rhodantha were used as objects to identify the characters and microscopic characteristics of its fresh and dried medicinal materials.Mangiferin,water,total ash and extract contents were determined.The similarity of the whole chemical components was evaluated synchronously by HPLC fingerprint.Results:There were significant differences in plant height,branch number,leaf length and leaf width between wild and cultivated Gentiana rhodantha.The plant height of wild Gentiana rhodantha was 46.80 to 80.93 cm,which was 1.3 times that of cultivated Gentiana rhodantha.The internode length and leaf area of wild Gentiana rhodantha were 1.24 times and 1.17 times that of cultivated herbs,respectively.However,the number of secondary branches of cultivated Gentiana rhodantha was 1.59 times that of wild Gentiana rhodantha.There was no significant difference between the microscopic characteristics of wild and cultivated Gentiana rhodantha.Taking mangiferin as the index component,the mangiferin content of cultivated Gentiana rhodantha was 36.76 to 50.70 mg/g,and that of wild Gentiana rhodantha was 38.28 to 51.62 mg/g,which met the requirements of the 2020 edition of Chinese pharmacopoeia.A total of 18 common peaks were identified by HPLC fingerprint,and the similarity was above 0.900.Conclusion:There is no significant difference between wild and cultivated Gentiana rhodantha except plant height and number of branches,which indicates that the quality of wild and cultivated Gentiana rhodantha is similar.In the future,comparative studies on pharmacodynamics should be carried out to ensure the satefy and effectiveness of clinical drugs.
