基于UPLC-QTOF-MS的白花蛇舌草醇提物成分表征及其抗肝癌谱效关系

Characterization of the compounds of Hedyotis diffusa alcohol extract based on UPLC-QTOF-MS and its spectral effect relationship of liver cancer

摘要 目的通过超高效液相色谱四极杆飞行时间串联质谱(UPLC-QTOF-MS)技术及谱效关联性分析阐明白花蛇舌草醇提物抗肝癌物质基础。方法高效液相色谱(HPLC)法建立10批白花蛇舌草醇提物指纹图谱,以白花蛇舌草醇提物对肝癌细胞SMMC-7721的增殖抑制率和凋亡坏死率为指标,分析白花蛇舌草醇提物抗肝癌的药效。再分别以灰色关联法和偏最小二乘分析法分析白花蛇舌草醇提物抗肝癌的谱效相关性。最后通过UPLC-QTOF-MS技术对共有峰进行鉴定。结果白花蛇舌草醇提物可抑制肝癌细胞SMMC-7721的增殖并诱导其凋亡,抑制增殖的IC_(50)为3.42 mg·mL^(-1)。谱效分析表明12、4、44、9、11、7、10号色谱峰呈正相关,2、38、27、29号色谱峰呈负相关。其中12号峰为5-咖啡酰奎宁酸,4号峰为4-OD-吡喃葡萄糖基-对香豆酸,9号峰为tachiogroside B,2号峰为丁二酸。结论白花蛇舌草醇提物能够抑制肝癌细胞的增殖,其物质基础可能为5-咖啡酰奎宁酸、4-OD-吡喃葡萄糖基-对香豆酸、tachiogroside B等。 Objective To elucidate the substance basis of Hedyotis diffusa alcohol extract for anti liver cancer through UPLC-QTOF-MS technology and spectral correlation analysis.Methods HPLC was used to establish ten batches of fingerprint spectra of Hedyotis diffusa alcohol extract,and the anti liver cancer efficacy of Hedyotis diffusa alcohol extract was analyzed using the proliferation inhibition rate and apoptosis necrosis rate of liver cancer cell line SMMC-7721 as indicators.Further analyze the spectrum effect correlation of Hedyotis diffusa alcohol extract against liver cancer using grey relational analysis and partial least squares analysis.Finally,UPLC-QTOF-MS technology was used to identify the common peaks.Results Hedyotis diffusa alcohol extract significantly inhibited the proliferation and induced apoptosis of liver cancer cell line SMMC-7721,with an IC_(50) of 3.42 mg·mL^(-1).Spectral analysis shows that peaks 12,4,44,9,11,7,and 10 are positively correlated,while peaks 2,38,27,and 29 are negatively correlated.Among them,peak 12 is 5-caffeoylquinic acid,peak 4 is 4-OD-p-glucopyranosyl-p-coumaric acid,peak 9 is tachiogroside B,and peak 2 is succinic acid.Conclusion The Hedyotis diffusa alcohol extract could inhibit the proliferation of liver cancer cells,and its material basis may be 5-caffeoylquinic acid,4-OD-glucopyranosyl-p-coumaric acid,tachiogroside B,etc.
