橙皮素通过调控P53通路关键蛋白抑制肝癌细胞SMMC-7721增殖、迁移并诱导凋亡

Hesperidin inhibits the proliferation,migration and induced apoptosis in SMMC-7721 through regulating the key protein of P53 pathway

摘要 目的 以人正常肝细胞L02和肝癌细胞SMMC-7721为研究对象,探讨橙皮素(Hesperetin,Hst)抑制肝癌细胞的增殖和迁移,并探究其具体机制。方法 通过使用60和120μmol/L的Hst处理L02和SMMC-7721细胞,采用cell counting kit-8(cck-8)法检测细胞增殖情况;划痕实验、transwell小室检测迁移能力和侵袭情况;流式细胞仪检测细胞凋亡情况;Western blot检测p53蛋白(p53,tumor protein 53)、细胞周期依赖性蛋白激酶抑制因子1A(p21,cyclin-dependent kinase inhibitor 1A)与Bcl2关联X蛋白(Bax,Bcl-2 associated X protein)的表达情况,对比Hst对人正常肝细胞L02与肝癌细胞SMMC-7721的影响。结果 与溶剂对照组相比,随着Hst浓度的升高,SMMC-7721细胞存活率、迁移能力和侵袭情况明显下降和减弱(P<0.05),Hst在120μmol/L时细胞凋亡率升高(F=6.33,P<0.05),在120μmol/L浓度下能够上调Bax(F=6.42,P<0.05)并下调p53(F=4.78,P<0.05)和p21(F=5.58,P<0.05)蛋白的表达水平;而对正常肝细胞的增殖、迁移、细胞凋亡率与蛋白表达影响不明显(P> 0.05)。结论 Hst能够抑制肝癌细胞SMMC-7721的增殖、迁移与侵袭,促进癌细胞凋亡,调控机制与p53通路相关。 Objective This study utilized the human hepatic cell line L02 and human hepatocellular carcinoma cells SMMC-7721 as the test subjects to investigate the inhibitory effects of Hesperetin(Hst)on the viability and migration of liver cancer cells,and explored its specific mechanism.Methods The L02 and SMMC-7721 cells were treated with Hst at the concentrations of 0,60,and 120μmol/L.Cell counting kit 8(CCK)-8 was used to detect the cells viability.The wound healing and transwell test were used to detect migration capacity and invasion.The apoptosis was detected by flow cytometry.Western blot method was used to detect the expression of p53 protein(p53),cyclin-dependent kinase inhibitor 1A(p21)and Bcl-2 associated X protein(Bax).Compared the experimental result of human normal liver cells and SMMC-7721 cells groups.Results Compared with the solvent control group,the augmentation of Hst concentration resulted in a significant reduction(P<0.05)in the viability,migration,and invasion of SMMC-7721 cells(P<0.05).Hst increased the apoptosis rate at 120μmol/L(F=6.33,P<0.05).At the concentration of 120μmol/L,Hst up-regulated the expression of Bax(F=6.42,P<0.05)and down-regulated the expression of p53(F=4.78,P<0.05)and p21 protein(F=5.58,P<0.05).There was no significant effect on the viability,migration,and protein expression of L02 cells(P>0.05).Conclusion Hst can inhibit the viability,migration,and invasion of SMMC-7721 cells,promote apoptosis of cancer cells,and the regulatory mechanism is related to p53 pathway.
