聚苯乙烯纳米塑料对东海原甲藻的毒性效应

摘要 针对纳米塑料对不同海洋微藻的毒性效应差异及其作用机制尚未明确的问题,本文从细胞和分子层面分析了不同浓度(0.1、1和10 mg/L)的聚苯乙烯纳米塑料(Polystyrene nanoplastics,PS-NPs)对东海原甲藻(Prorocentrum donghaiense)的毒性作用。结果显示,10 mg/L的PS-NPs对东海原甲藻的毒性效应大于0.1和1 mg/L的PS-NPs。在细胞层面,PS-NPs抑制了东海原甲藻的生长,藻细胞产生了氧化应激反应,活性氧(Reactive oxygen species,ROS)水平上升,丙二醛(Malondialdehyde,MDA)含量增加,超氧化物歧化酶(Superoxide dismutase,SOD)和过氧化氢酶(Catalase,CAT)活性升高,细胞凋亡率增高。在分子层面,经过PS-NPs暴露处理后的东海原甲藻,其核糖体、蛋白酶体和内质网蛋白质加工相关的基因表达发生了变化:氧化磷酸化过程中相关基因表达量上调;为了应对氧化应激,过氧化物酶体过程中的相关基因表达量同样上调。另外,光合作用天线蛋白通路中的相关基因表达量下调,导致藻细胞的光捕获能力下降。本研究可为进一步评估纳米塑料对海洋微藻的毒性效应提供科学依据。 In this paper,the toxic effects of different concentrations(0.1,1,10 mg/L)of polystyrene nanoplastics(PS-NPs)on Prorocentrum donghaiense were analyzed at both cellular and molecular levels.The results showed that the toxic effects of 10 mg/L PS-NPs on P.donghaiense were greater than those of 0.1 and 1 mg/L.At the cellular level,PS-NPs inhibited the growth of P.donghaiense,the cells responded to oxidative stress,the level of reactive oxygen species(ROS)increased,the content of malondialdehyde(MDA)increased,the activities of superoxide dismutase(SOD)and catalase(CAT)increased,and the apoptosis rate of cells increased.At the molecular level,the expression of genes related to ribosome,proteasome and protein processing in endoplasmic reticulum was changed on P.donghaiense when PS-NPs were put into the culture medium.The expression of related genes in the oxidative phosphorylation process was up-regulated in the PS-NPs group,and in response to oxidative stress,the expression of related genes in the peroxisome was up-regulated as well.In addition,the expression of related genes in the photosynthesis-antenna proteins was down-regulated,and the light-harvesting capacity of cells decreased.This study can provide scientific basis for further evaluating the toxic effects of nanoplastics on marine microalgae.
