基于线粒体DNA控制区序列的养殖与野生褐菖鲉群体遗传多样性比较分析

Comparative Analysis of Genetic Variation in Cultured and Wild Populations of Sebastiscus marmoratus Based on Mitochondrial DNA Control Region Sequence

摘要 褐菖鲉是近岸岛礁海域广泛分布的岩礁生境底栖鱼类,也是我国东海区重要的增殖放流种。旨在通过分析线粒体DNA控制区(D-loop)序列,探究养殖与野生褐菖鲉群体在遗传多样性及遗传分化水平方面的差异。结果显示,养殖与野生褐菖鲉DNA序列的A+T含量分别为56.75%和51.56%。养殖群体的单倍型多样性(hd=0.800±0.057)、核苷酸多样性(π=0.01779±0.00141)明显低于野生群体的单倍型多样性(hd=0.957±0.026)、核苷酸多样性(π=0.02204±0.00404),表明褐菖鲉养殖群体的遗传多样性较低。系统发育树、单倍型最小跨度树及遗传分化指数显示,野生和养殖群体间遗传分化水平较低。经过对单倍型最小跨度树和系统发育树的分析,并未检测到明显的谱系结构存在。研究结果表明,在褐菖鲉人工增殖苗种繁育过程中,应通过增加亲本群体来源提高人工增殖苗种的单倍型多样性,以降低养殖放流群体与野生群体之间在遗传多样性上的差异。 The marbled rockfish(Sebastiscus marmoratus)is an ecologically and economically important inshore species in eastern Asia.In recent years,it has become one of the key species for aquaculture-based stock enhancement in the East China Sea.This study investigates the genetic diversity of both cultured and wild populations of marbled rockfish through an analysis of the mitochondrial DNA control region(D-loop).Our findings reveal that the A+T content in cultured and wild populations is 56.75%and 51.56%,respectively.Comparative analysis shows that the haplotype diversity of the cultured population(hd=0.800±0.057)is significantly lower than that of the wild population(hd=0.957±0.026).Furthermore,the nucleotide diversity of the cultured population(π=0.01779±0.00141)is lower than that of the wild population(π=0.02204±0.00404),indicating a marked reduction in genetic diversity within the cultured marbled rockfish.Phylogenetic analysis,haplotype network evolutionary relationships,and genetic differentiation indices collectively suggest that there is no significant genetic differentiation between the cultured and wild populations.Our results highlight the importance of enhancing haplotype diversity during the seedling production process by broadening the parental population base of marbled rockfish.
