摘要
针对广东、海南省4个不同地方(阳江、湛江、榆林、琼海)网箱养殖的紫红笛鲷(Lutjanusargentimaculatus)群体和在三亚采集的野生紫红笛鲷群体,利用AFLP分子标记技术分析我国南方养殖的紫红笛鲷的遗传多态性。最适合的AFLP引物组合是E+AGC/M+CAA,扩增出的AFLP特征位点为107,养殖样品扩增出的AFLP的带数在54—74之间,野生样品扩增出的AFLP的带数为63,4个养殖点紫红笛鲷的多态性标记百分率都超过了40.00%,呈现一定程度的遗传多样性。总的来讲,4个养殖地的紫红笛鲷群体之间的遗传距离差异较小,但与野生群体的遗传距离相对较大,UPGMA聚类分析明显将养殖群体与野生群体分开,表明紫红笛鲷养殖群体已经与野生群体存在一定程度的遗传隔阂。
The genetic diversities of Lutjanus argentimaculatus (Forsskl) of the stocks at 4 mariculture spots in Guangdong & Hainan provinces and the wild stock in Sanya were analyzed by amplified fragment length polymorphism (AFLP) molecular technique. The optimal AFLP pattern was obtained, with a primer combination of E+AGC/M+CAA selective nucleotides. There were totally 107 AFLP loci in this study. The number of AFLP markers fluctuated between 54 and 74 for the maricultural stocks,and was 63 for the wild stock. The percentage of polymorphic markers of each maricultural stock was over 40.00%, showing a high genetic polymorphism to a certain extent. In general,the distinction of pairwise genetic distances among maricultural stocks was unobvious,whereas the difference of pairwise genetic distance between the maricultural stocks and the wild stock was comparatively large. In UPGMA dendrogram,the maricultural stocks were clustered together,apart from the wild stock, indicating that inbreeding had caused the genetic estrangement between the maricultural stocks and the wild stock.
