人工老化大豆种子的幼苗生理特性及DNA甲基化变异 被引量:1

Physiological Characteristics and DNA Methylation Variation of Seedlings from Artificially Aged Soybean Seeds

摘要 为了探究老化种子对幼苗的生理状态和表观遗传的影响,选用2个大豆品种“吉农43”和“吉农36号”作为试验材料,在(55±1)℃相对湿度(RH)100%条件下,人工老化处理种子5,10,15 min,以未处理种子作为对照,测定幼苗的生理指标和分析DNA的5'-CCGG位点的甲基化变异。结果表明:随老化时间延长,老化大豆种子幼苗的过氧化物酶(POD)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性表现为下降趋势,当老化15 min时,两品种的POD活性极显著地低于ck 32.9%和14.5%,SOD的活性分别比ck降低了16.4%和22.2%,CAT的活性则降低了9.3%和9.1%。老化大豆种子幼苗的MDA含量则随老化时间延长表现为升高趋势,老化15 min时,“吉农43”和“吉农36号”的MDA含量分别是2.92μmol/g和3.23μmol/g。老化种子15 min时,DNA的5'-CCGG位点单链CHG半甲基化和双链CG全甲基化水平随老化程度加深呈升高趋势,而内外胞嘧啶同全甲基化水平则表现为降低。研究结果表明人工老化大豆种子会降低生理活性酶的活性,对DNA的5'-CCGG位点的甲基化总体水平改变较小,但不同模式的甲基化水平因老化而改变较大且表现为不同变化趋势。 In order to explore the effects of aged seeds on physiological state and epigenetic inheritance of seedlings,two soybean varieties Jinong 43 and Jinong 36 were selected as experimental materials.Seeds were artificially aged at(55±1)℃ with relative humidity(RH)100%for 5,10 and 15 min,and untreated seeds were used as control.Physiological indexes of seedlings were determined and DNA methylation variation at the 5'-CCGG site was analyzed.The results showed that with the extension of aging time,the activities of peroxidase(POD),superoxide dismutase(SOD)and catalase(CAT)of the aged soybean seeds showed a decreasing trend.When aging for 15 min,POD activity of the two cultivars was significantly lower than ck,by 32.9%and 14.5%,SOD activity was decreased by 16.4%and 22.2%,and CAT activity was decreased by ck 9.3%and 9.1%.The MDA content of aging soybean seeds increased with aging time,and the MDA content of“Jinong 43”and“Jinong 36”were 2.92μmol/g and 3.23μmol/g,respectively,after aging for 15 min.When the seeds were aged for 15 min,the 5'-CCGG DNA hemimethylation levels of single CHG and double stranded CG were increased with the aging degree,while the endogenous and exogenous cytosine methylation levels were decreased.The results indicated that artificial aging of soybean seeds could reduce the activity of physiologically active enzymes and change the overall methylation level of DNA 5'-CCGG site slightly,but the methylation level of different patterns changed greatly and showed different trends due to aging.
