降解铁皮石斛多糖的内生真菌筛选及产物组分分析 被引量:1

摘要 目的:以铁皮石斛多糖为唯一营养物质,从内生真菌中筛选可降解多糖的潜力菌株。方法:利用微生物液体培养发酵技术筛选可降解铁皮石斛粗多糖的内生真菌,分别采用高效液相色谱法、苯酚硫酸法、二喹啉甲酸(BCA)法、间羟基联苯法和1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化法测定降解产物的相对分子质量、总糖含量、总蛋白质含量、糖醛酸含量和单糖组成;对降解产物的紫外光谱图、红外光谱图、表观结构进行比较和分析;采用1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除实验评价降解产物的抗氧化能力。结果:发现4株内生真菌可降解铁皮石斛粗多糖DOP-60%。其中,D-25菌株的降解产物相对分子质量最小(2.55×10^(3)),得率最高(76.7%);DOP-60%与降解产物CDOP-3~CDOP-6的单糖组成类型相同,甘露糖含量增加。抗氧化活性研究表明,降解产物中CDOP-5的DPPH自由基清除能力相对较强。结论:利用铁皮石斛内生真菌降解其多糖,得到4株可降解铁皮石斛多糖的潜力菌株,菌株D-25降解效果最好、多糖得率最高。 Objective:To screen potential strains of degradable polysaccharides from endophytic fungi using Dendrobium officinale polysaccharides(DOP)as the sole nutrient.Methods:Endophytic fungi capable of degrading crude DOP were screened using microbial liquid culture fermentation technology.The relative molecular weight,total sugar content,total protein content,uronic acid content,and monosaccharide composition of the degradation products were determined using high-performance liquid chromatography,phenol-sulfuric acid method,BCA method,m-hydroxybiphenyl method,and PMP pre-column derivatization method.The ultraviolet spectrum,infrared spectrum,and apparent structure of the degradation products were compared and analyzed.The antioxidant capacity of the degradation products was evaluated by 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radical scavenging assay.Results:Four endophytic fungi were found to degrade crude DOP-60%.Among them,the degradation product of strain D-25 had the smallest relative molecular mass(2.55×10^(3))and the highest yield(76.7%).The monosaccharide composition of DOP-60%and the degradation products CDOP-3 to CDOP-6 were the same,with an increase in mannose content.Antioxidant activity studies showed that the DPPH free radical scavenging ability of CDOP-5 in the degradation products was relatively strong.Conclusion:For the first time,endophytic fungi from D.officinale were used to degrade its polysaccharides,yielding four potential strains capable of degrading DOP.Strain D-25 exhibited the best degradation effect and the highest polysaccharide yield.
