经筋恢刺联合中药熏药治疗对膝骨关节炎模型大鼠NF-κB信号通路的影响 被引量:1

Effect of Meridian Sinew Lateral Needling Combined with Chinese Medical Fumigation on NF-κB Signaling Pathway in KOA Model Rats

摘要 目的:探讨经筋恢刺法联合中药熏药治疗膝骨关节炎(KOA)的作用机制及对NF-κB信号通路的影响。方法:碘乙酸钠诱导大鼠KOA模型,随机分为正常组(NC组)、模型组(M组)、对照组(DZ组)、经筋恢刺组(HC组)与经筋恢刺联合中药熏药组(LH组);治疗前后进行行为学评分,治疗后取膝关节液检测IL-1β、IL-6与TNF-α,取膝关节组织进行病理组织学观察,PCR法检测IKK mRNA、NF-κB p65 mRNA相对表达量,Western blot法检测蛋白表达。结果:治疗后,与M组比较,各治疗组行为学评分均显著降低,其中LH组评分显著低于DZ组,差异具有统计学意义(P<0.05);各治疗组IL-1β水平均显著降低,LH组IL-6水平显著降低,差异具有统计学意义(P<0.05),TNF-α差异无统计学意义(P<0.05);各治疗组组织破坏均减轻,其中LH组减轻程度最好;各治疗组IKK mRNA、NF-κB p65 mRNA相对表达量及p-IKK/IKK、p-p65/p65水平均降低,其中LH组p-IKK/IKK、p-p65/p65水平低于DZ组,差异具有统计学意义(P<0.05)。结论:经筋恢刺法联合中药熏药治疗可提高模型大鼠患肢行为学评分、改善关节组织破坏、降低炎症因子水平及抑制NF-κB信号通路,从而改善局部炎症反应。 Objective:To explore the mechanism of meridian sinew lateral needling combined with Chinese medical fumigation in treatment of knee osteoarthritis(KOA)and its impacts on NF-κB signaling pathway.Methods:The rats were randomly divided into the normal control group(NC),the model(M)group,the Chinese medication control(DZ)group,the meridian sinew lateral needling(HC)group,and the meridian sinew lateral needling combined with Chinese medical fumigation(LH)group.The KOA model was elaborated by using Sodium Iodoacetate.Behavioral scoring was assessed before and after the treatment.Knee joint fluid was collected after the treatment for testing the levels of IL-1β,IL-6,and TNF-α.Knee joint tissues were collected for histopathological observation.Relative mRNA expressions of IKK NF-κB and p65 were detected by using PCR,and their protein expressions were detected by applying Western blot.Results:The behavioral scores were lower in all treatment groups than that in the M group after the treatment,among which the score in the LH group was lower than that in the DZ group.The levels of IL-1βwere decreased in all treatment groups,the level of IL-6 was decreased in the LH group,but the levels of TNF-αdid not show significant changes.The levels of IL-1 were significantly reduced in each treatment group,with a significant decrease in the LH group.The tissue damage in each treatment group was reduced,with the LH group showing the best degree of reduction.The relative mRNA expressions of IKK NF-κB and p65,as well as the values of p-IKK/IKK and p-p65/p65 were decreased in each treatment group;among which,the values of p-IKK/IKK and p-p65/p65 in the LH group were lower than those in the DZ group.Conclusion:The regimen of meridian sinew lateral needling combined with Chinese medical fumigation can improve the behavioral score of the affected limbs in the model rats,it can improve joint tissue damage,reduce inflammatory factor levels,and inhibit NF-κB signaling pathway,thus to improve local inflammatory response.
