摘要
旨在探讨猪肺炎支原体(Mhp)感染3D4/21细胞后引起的活性氧(ROS)分泌变化及其相关的基因表达情况,并通过时间点的选择来分析表达发生改变基因的功能和潜在的感染机制。采用高通量测序技术,分析了Mhp感染细胞后的基因表达情况,并对差异表达基因(DEGs)进行了基因本体(GO)和京都基因与基因组百科全书(KEGG)功能富集分析。结果:Mhp感染细胞和未感染细胞之间共有1940个DEGs,其中1070个上调,871个下调;GO功能分析发现Mhp对3D4/21细胞的影响主要集中在细胞进程、代谢进程、生物调节等方面;KEGG富集分析发现差异基因主要富集在癌症的发生发展、氧化还原、化学致癌、丝裂原活化蛋白激酶(MAPK)和磷脂酰肌醇3-激酶-蛋白激酶B(PI3K-Akt)等相关信号通路。本研究在分子水平上为Mhp感染机制的理解提供新的视角,并为后期防治猪支原体肺炎的靶向药物研发提供了初步研究基础。
The study was to investigate the changes in ROS secretion and related gene differential expression following Mycoplasma hyo⁃pneumoniae(Mhp)infection of 3D4/21 cells,and to analyze the functions and potential infection mechanisms of these differentially ex⁃pressed genes by selecting appropriate time points.High-throughput sequencing technology was used to analyze gene expression in Mhp-in⁃fected cells,and functional enrichment analysis was performed on the differentially expressed genes(DEGs)using GO and KEGG.The results showed that there were 1940 differentially expressed genes between Mhp-infected cells and blank control cells,with 1070 upregulat⁃ed and 871 downregulated.The GO functional analysis revealed that the impact of Mhp on 3D4/21 cells primarily focused on cellular proces⁃ses,metabolic processes,and biological regulation.The KEGG enrichment analysis indicated that the differentially expressed genes were mainly enriched in pathways related to cancer development,redox reactions,chemical carcinogenesis,MAPK signaling,and PI3K-Akt sig⁃naling.This study provided new insights into the molecular mechanisms of Mhp infection and laid a foundation for the development of targeted drugs for the prevention and treatment of Mycoplasmal pneumonia in swine.
