鹿皮胶特征肽的发现及其在鹿角胶中鹿皮源成分检测上的应用 被引量:1

摘要 目的寻找可以区分鹿皮胶、鹿角胶两种不同来源动物胶的特征肽,并建立鹿角胶中掺杂鹿皮源成分的检测方法。方法利用胰蛋白酶对鹿皮胶和鹿角胶样品进行酶切,使用超高效液相色谱-三重四级杆质谱联用仪(UPLC-QQQ/MS)分别测定鹿皮胶和鹿角胶的肽图,对肽图进行比对分析,找出在鹿皮胶中响应强但在鹿角胶中没有响应或响应极弱的分子离子,并对这些分子离子建立MRM方法,通过专属性验证得到两个鹿皮胶特征肽。将这两个鹿皮胶特征肽作为鹿皮胶的鉴别特征,对鹿角胶样品进行测定。结果在鹿皮胶样品中检出了新发现的两个鹿皮胶特征肽,在鹿角胶、阿胶、猪皮胶、牛皮胶、马皮胶、羊皮胶、骆驼皮胶、龟甲胶、鳖甲胶样品中均未检出这两个鹿皮胶特征肽。该方法的检出限度为含0.2%鹿皮胶的鹿角胶样品。对12批鹿角胶样品进行检测,在1批样品中检出了鹿皮胶特征肽。结论所建立的方法经验证,专属性强、灵敏度高,可用于鹿皮胶的鉴别及鹿角胶中鹿皮源成分的检测。 OBJECTIVE Searching for specific peptides that can distinguish between deer-hide gelatin and deer-antler gelatin,and establish a detection method for deer-hide gelatin mixed in deer-antler gelatin.METHODS Using trypsin to cleave samples of deer-hide gelatin and deer-antler gelatin,the peptide chromatogram of deer-hide gelatin and deer-antler gelatin were determined using ultra-high performance liquid chromatography triple quadrupole mass spectrometry(UPLC-QQQ/MS).The peptide chromatogram were compared and analyzed to identify the exclusive molecular ions of deer-hide gelatin,and an MRM method was established for these molecular ions,Two deer-hide gelatin specific peptides were obtained through specificity verification.Use these two deer-hide gelatin specific peptides as identification features for deer-hide gelatin and measure the deer-antler gelatin samples.RESULTS Two new specific peptides of deer-hide gelatin were detected in the samples of deer-hide gelatin,but they were not detected in the samples of deer-antler gelatin,ass-hide gelatin,pig-hide gelatin,cattle-hide gelatin,horse-hide gelatin,sheep-hide gelatin,camel-hide gelatin,tortoise-shell gelatin and turtle-shell gelatin.The detection limit of this method is for deer-antler gelatin samples containing 0.2%deer-hide gelatin.Twelve batches of deer-antler gelatin samples were tested,and deer-hide gelatin specific peptides were detected in one batch of samples.CONCLUSION The established method has been validated to have strong specificity and high sensitivity,and can be used for the identification of deer-hide gelatin and the detection of deer-hide gelatin mixed in deer-antler gelatin.
