摘要
目的:基于Toll样受体4(TLR4)/髓样分化蛋白88(MyD88)/核因子-κB(NF-κB)信号通路探讨明君散对动脉粥样硬化(AS)大鼠的影响。方法:采用高脂饲料联合维生素D3法建立大鼠AS模型,分为空白组、模型组、西药组[阿托伐他汀1 mg(/kg·d)]、明君散低剂量组[2.35 mg(/kg·d)]、明君散中剂量组[4.70 mg(/kg·d)]、明君散高剂量组[9.40 mg(/kg·d)],每组10只。灌胃给药8周后,检测血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平;采用苏木精-伊红(HE)染色观察大鼠胸主动脉组织形态学变化;实时荧光定量聚合酶链式反应(RT-PCR)检测大鼠主动脉TLR4、MyD88、NF-κB mRNA表达水平;蛋白免疫印迹法(Western Blot)检测大鼠主动脉TLR4、MyD88、NF-κB蛋白表达水平。结果:与模型组比较,西药组和明君散中剂量组、明君散高剂量组血清TC、TG,LDL-C水平降低,HDL-C水平升高(P<0.05或P<0.01);西药组和明君散各剂量组大鼠主动脉组织中TLR4、MyD88、NF-κB mRNA和蛋白表达水平均降低(P<0.05或P<0.01)。结论:明君散可能通过抑制TLR4/MyD88/NF-κB信号通路发挥抗动脉粥样硬化作用。
Objective:To explore the effect of Mingjun Powder on atherosclerotic rats based on the Toll-like receptor 4(TLR4)/myeloid differentiation protein 88(MyD88)/nuclear factor-κB(NF-κB)signaling pathway.Methods:The atherosclerotic model of rats was established by high-fat feed combined with vitamin D3 method.The rats were divided into the blank group,the model group,the Western medicine group[atorvastatin 1 mg/(kg·d)],the low-dose Mingjun Powder group[2.35 mg/(kg·d)],the medium-dose Mingjun Powder group[4.70 mg/(kg·d)],and the high-dose Mingjun Powder group[9.40 mg/(kg·d)],with 10 rats in each group.After gavaging for eight weeks,the levels of serum total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)were detected.Hematoxylin-eosin(HE)staining was used to observe the histological morphological changes of the thoracic aorta in rats.The expressions of TLR4,MyD88 and NF-κB mRNA in the aorta of rats were detected by reverse transcription polymerase chain reaction(RT-PCR).The expressions of TLR4,MyD88,and NF-κB proteins in the aorta of rats were detected by Western Blot.Results:Compared with the model group,the levels of serum TC,TG,and LDL-C in the Western medicine group,the medium-dose Mingjun Powder group,and the high-dose Mingjun Powder group decreased,and the level of HDL-C increased(P<0.05 or P<0.01).The expressions of TLR4,MyD88,NF-κB mRNA and protein in the aortic tissues of rats in the Western medicine group and each dose Mingjun Powder groups all decreased(P<0.05 or P<0.01).Conclusion:Mingjun Powder might play an anti-atherosclerotic effect by inhibiting the TLR4/MyD88/NF-κB signaling pathway.
