芜菁纤维提取工艺及抗氧化研究

Extraction of dietary fiber from Brassica rapa L.and its antioxidant capacity

摘要 为了优化芜菁纤维及膳食纤维的提取工艺,测定可溶性膳食纤维抗氧化活性。将芜箐粗纤维采用酸性洗涤法提取,膳食纤维采用碱法提取。以温度、碱液浓度、碱解时间为因素设计正交试验。通过测定芜菁可溶性膳食纤维对DPPH自由基、羟基自由基、ABTS^(+)自由基的清除和对三价铁离子的还原能力来评价其抗氧化效果。结果显示各组分含量分别为:芜菁粗纤维0.7%,不可溶性膳食纤维2.24%,可溶性膳食纤维1.67%,总膳食纤维3.91%。芜菁膳食纤维最优提取条件为温度70℃、碱解时间80 min、碱液浓度9%。芜菁可溶性膳食纤维对DPPH自由基、羟基自由基、ABTS^(+)自由基的清除能力和对三价铁离子的还原能力与浓度呈正相关,三种自由基清除率最高分别可达30.50%、79.32%、85.83%。本研究从芜菁中提取的粗纤维和膳食纤维含量高,芜菁可溶性膳食纤维有着一定的抗氧化效果。 In order to optimize the extraction process of fiber from Brassica rapa L.(turnip)and to evaluate the soluble dietary fiber antioxidant activity;crude fiber of turnip was extracted using acid treatment,while dietary fiber was extracted using an alkaline method.An orthogonal experimental design was employed to optimize the alkaline extraction,with temperature,alkali concentration,and extraction time as factors.The antioxidant activity of the soluble dietary fiber fraction was assessed by its ability to scavenge DPPH radicals,hydroxyl radicals,and ABTS^(+)radicals,as well as its ferric reducing antioxidant power.The extracted fiber content was as follows:turnip crude fiber 0.7%,insoluble dietary fiber 2.24%,soluble dietary fiber 1.67%,and total dietary fiber 3.91%.The optimal extraction conditions for turnip dietary fiber were determined to be 70℃,80 min extraction time,and 9%alkali concentration.The turnip soluble dietary fiber exhibited significant antioxidant activity,with scavenging rates of 30.50% for DPPH radicals,79.32% for hydroxyl radicals,and 85.83% for ABTS^(+)radicals.All three free radical scavenging rates and ferric reducing antioxidant power showed concentration-dependent relationships.This study demonstrated that the crude fiber and dietary fiber extracted from turnip contained high fiber content,and the soluble dietary fiber exhibited certain antioxidant activity.
