摘要
目的观察葛根芩连汤对肠源性内毒素血症(IETM)大鼠细胞凋亡与细胞增殖的影响。方法采用细胞增殖检测分析法检测不同剂量葛根芩连汤对SW480细胞增殖活性的抑制作用;采用实时荧光定量聚合酶链式反应、蛋白免疫印迹法及免疫组化法检测不同剂量葛根芩连汤对大鼠肠黏膜增殖细胞核抗原(PCNA)、B淋巴细胞瘤-2(BCL-2)和BCL-2相关X蛋白(Bax)mRNA及蛋白表达;采用TUNEL染色法检测大鼠肠黏膜细胞凋亡的情况。结果与空白对照组比较,模型对照组细胞存活率明显降低;与模型对照组比较,除低剂量组外,葛根芩连汤均能提高其他剂量组细胞存活率,且呈现出剂量依赖性,差异有统计学意义(P<0.05)。与空白对照组比较,模型对照组PCNA和BCL-2 mRNA表达降低,Bax mRNA表达升高;与模型对照组比较,葛根芩连汤不同剂量组与阳性药物对照组PCNA和BCL-2 mRNA表达均升高,Bax mRNA表达均降低,具有一定的剂量依赖性,差异有统计学意义(P<0.05)。蛋白免疫印迹法结果和免疫组化结果也呈现一致的趋势。与空白对照组比较,模型对照组凋亡水平最高;随着葛根芩连汤剂量的提高,凋亡水平减弱,呈现出剂量依赖性,差异有统计学意义(P<0.05)。结论葛根芩连汤通过上调PCNA和BCL-2表达、下调Bax表达,从而抑制IETM大鼠细胞凋亡并促进细胞增殖。
Objective The study aims to observe the effect of Gegen Qinlian Decoction on apoptosis and proliferation of intestinal endotoxemia(IETM)rats.Methods The cell proliferation assay was used to detect the inhibitory effect of different doses of Gegen Qinlian Decoction on the proliferation of SW480 cells.The expressions of proliferating cell nuclear antigen(PCNA),B-cell lymphoma-2(BCL-2)and BCL-2-associated X protein(Bax)mRNA and protein in intestinal mucosa of rats treated with different doses of Gegen Qinlian Decoction were detected by real-time fluorescent quantitative polymerase chain reaction,Western blot and immunohistochemistry.TUNEL staining was used to detect the apoptosis of intestinal mucosal cells.Results Compared with the blank control group,the cell survival rate of the model control group was significantly decreased.Compared with the model control group,Gegen Qinlian decoction could improve the cell survival rate of all the other dose groups except the low dose group,and the difference was statistically significant(P<0.05).Compared with the blank control group,the expression of PCNA and BCL-2 mRNA in the model control group decreased,and the expression of Bax mRNA increased.Compared with the model control group,the expressions of PCNA and BCL-2 mRNA in all dose groups of Gegen Qinlian Decoction and positive drug control group were increased,and the expression of Bax mRNA was decreased,and the effect of each dose group was dose-dependent,the difference was statistically significant(P<0.05).The results of Western blot and immunohistochemistry also showed a consistent trend.Compared with the blank control group,the apoptosis level of the model control group was the highest.With the increase of the dose of Gegen Qinlian Decoction,the level of apoptosis decreased,showing a dose-dependent difference(P<0.05).Conclusion Gegen Qinlian Decoction can inhibit cell apoptosis and promote cell proliferation in IETM rats by up-regulating the expression of PCNA and BCL-2 and down-regulating the expression o f Bax.
