全蝎膏通过p38/MK2通路对糖尿病溃疡大鼠新生血管及创面炎症反应的影响

Effects of Scorpion Ointment(全蝎膏)on neovascularization and wound inflammation in diabetic ulcer rats via the p38/MK2 pathway

摘要 目的探讨与分析全蝎膏通过p38/MK2通路对糖尿病溃疡大鼠新生血管及创面炎症反应的影响。方法将糖尿病溃疡大鼠模型(n=27)随机平分为三组,即模型组、凡士林组、全蝎膏组,每组9只,三组分别给予外敷生理盐水纱条、凡士林纱条、全蝎膏纱条治疗,持续21d,检测大鼠新生血管及创面炎症反应变化情况。结果糖尿病溃疡大鼠模型造模成功后出现反应迟钝、行动迟缓、多尿等现象,凡士林组、全蝎膏组治疗第7天、第14天、第21天的体质量显著高于模型组(P<0.05),全蝎膏组也显著高于凡士林组(P<0.05);凡士林组、全蝎膏组治疗第7天、第14天、第21天的伤口愈合率显著高于模型组(P<0.05),全蝎膏组也显著高于凡士林组(P<0.05);凡士林组、全蝎膏组治疗第7天、第14天、第21天血清IL-1β、IL-6含量都显著低于模型组(P<0.05),全蝎膏组与凡士林组相比也显著降低(P<0.05);凡士林组、全蝎膏组的治疗第21天创面新生血管数量显著高于模型组(P<0.05),全蝎膏组与凡士林组相比显著提高(P<0.05);凡士林组、全蝎膏组的治疗第21天创面p38与MK2蛋白相对表达水平显著低于模型组(P<0.05),全蝎膏组显著低于凡士林组(P<0.05)。结论全蝎膏在糖尿病溃疡大鼠的应用能提高体质量与改善伤口愈合率,能促进新生血管形成,同时通过抑制p38与MK2蛋白的表达,抑制创面炎症反应,促进糖尿病溃疡创面愈合。 Objective To investigate and analyze the effects of Scorpion Ointment(全蝎膏,SO)on neovascularization and wound inflammation in diabetic ulcer(DU)rats through the p38/MK2 pathway.Methods The model rats of DU(n=27)were randomly divided into three groups:the model group,vaseline group,and SO group,with 9 rats in each group.The three groups were treated externally with normal saline gauze strips,vaseline gauze strips,and SO gauze strips for 21 days,and the changes of neovascularization and wound inflammation were detected.Results After successful establishment of the DU model,the rats showed slow response and action,polyuria and other phenomena.The body weights of the vaseline group and SO group on the 7th,14th and 21st day of treatment were significantly higher than that of the model group(P<0.05);the SO group was significantly higher than that of vaseline group(P<0.05).The wound healing rates of the vaseline group and SO group on the 7th,14th,and 21st day of treatment were significantly higher than those of the model group(P<0.05),and the SO group was also significantly higher than vaseline group(P<0.05).The serum levels of IL-1βand IL-6 in the vaseline group and SO group were significantly lower than those in the model group on the 7th,14th,and 21st day of treatment(P<0.05),and the SO group also showed significant decrease compared to the vaseline group(P<0.05).On the 21st day of treatment,the number of new blood vessels in the wound of the vaseline group and SO group was significantly higher than that of the model group(P<0.05),and the SO group showed a significant increase compared to the vaseline group(P<0.05).On the 21st day of treatment,the relative expression levels of p38 and MK2 proteins in the wound of the vaseline group and SO group were significantly lower than those in the model group(P<0.05),and the SO group was significantly lower than vaseline group(P<0.05).Conclusion The application of SO in DU rats can increase body weight,enhance wound healing rate,promote angiogenesis,and meanwhile inhibit wound inflammation and promote the healing of DU by inhibiting the expressions of p38 and MK2.
