黄连解毒汤对2型糖尿病模型大鼠肠道菌群结构的影响 被引量:2

Effect of Huanglian Jiedu Decoction(黄连解毒汤)on the intestinal microbiota structure in rats with type 2 diabetes mellitus

摘要 目的运用16S rRNA测序技术研究黄连解毒汤对2型糖尿病模型大鼠肠道菌群的影响。方法将60只SD大鼠随机分为正常组、模型组、黄连解毒汤高、中、低剂量组和二甲双胍组。采用高脂饮食+链脲佐菌素构建2型糖尿病模型,造模成功后黄连解毒汤各剂量组和二甲双胍组予相应药物灌胃,正常组和模型组予等体积生理盐水灌胃,连续给药14d后收集各组大鼠粪便,运用16S rRNA测序方法观察各组大鼠肠道菌群结构的变化。结果与正常组相比,模型组肠道菌群丰富度和多样性增高;与模型组相比,黄连解毒汤各剂量组肠道菌群丰富度和多样性降低,门水平上,黄连解毒汤增加厚壁菌门丰度并降低拟杆菌门、变形菌门、放线菌门和脱硫菌门丰度。属水平上,黄连解毒汤增加异芽孢杆菌、布劳特氏菌和乳酸杆菌属丰度。LEfSe分析结果显示黄连解毒汤差异标志物是布劳特氏菌属、异芽孢杆菌属、乳酸杆菌属。通过PICRUSt2功能预测发现黄连解毒汤主要影响碳水化合物代谢途径和氨基酸代谢途径。结论黄连解毒汤能够调节2型糖尿病大鼠肠道菌群结构,增加产短链脂肪酸的菌种和有益菌丰度,影响碳水化合物和氨基酸代谢途径。 Objective To study the effect of Huanglian Jiedu Decoction(黄连解毒汤,HJD)on the intestinal microbiota of rats with type 2 diabetes mellitus(T2DM)through 16S rRNA sequencing technology.Methods A total of 60 Sprague-Dawley(SD)rats were randomly divided into the normal group,the model group,the HJD high-dose(HJD-H),medium-dose(HJD-M)and low-dose(HJD-L)groups and the metformin group,and a T2DM model was constructed using high-fat diet+streptozotocin.When the model was established,the HJD-H,HJD-M and HJD-L groups and the meformin group were gavaged with corresponding drugs,and the normal group and the model group were equally gavaged with saline.After continuous drug administration for 14 days,the feces of each group were collected,and the changes in the intestinal microbiota structure were observed by 16S rRNA sequencing method.Results Compared with the normal group,the richness and diversity of intestinal microbiota in the model group increased;compared with the model group,the richness and diversity of intestinal microbiota in the HJD groups decreased.At the phylum level,HJD increased the abundance of Firmicutes and decreased the abundance of Bacteroidota,Proteobacteria,Actinobacteriota and Actinobacteriota;at the genus level,HJD increased the abundance of Allobaculum,Blautia and Lactobacillus.The results of LEfSe analysis showed that the differential markers of HJD were Blautia,Allobaculum and Lactobacillus.Through PICRUSt2 function prediction,it was found that HJD mainly affected carbohydrate metabolism pathway and amino acid metabolism pathway.Conclusion HJD can regulate the structure of intestinal microbiota in T2DM rats,increase the abundance of short-chain fatty acid-producing and beneficial bacteria,and affect the metabolism pathways of carbohydrates and amino acids.
