缺氧诱导因子1在消化道肿瘤生长中的研究进展

摘要 消化道肿瘤是一组与消化系统及其附属器官相关的肿瘤,严重危害着人类的健康与安全。缺氧诱导因子1(HIF-1)是一种随着细胞内氧浓度变化而调节基因表达的转录激活因子,其表达随细胞内氧浓度变化而调节,并通过激活基因转录在肿瘤细胞对缺氧的适应中发挥关键作用,这些基因转录调节多种生物过程,包括葡萄糖代谢和侵袭转移,因此HIF-1成为有吸引力的消化道肿瘤治疗靶点。本文开展了HIF-1在消化道肿瘤生长进程中分别对细胞糖代谢、肿瘤转移和靶向治疗产生影响的综述。 Digestive tract malignancies,a heterogeneous group of tumors affecting the gastrointestinal system and associated organs,pose significant threats to global health.Hypoxia-inducible factor 1(HIF-1),an oxygen-sensitive transcriptional activator,dynamically regulates gene expression in response to intracellular oxygen fluctuations.This master regulator drives tumor cell adaptation to hypoxic microenvironments through transcriptional control of critical biological processes,including glycolytic reprogramming and metastatic progression,thereby establishing HIF-1 as an attractive target for gastrointestinal cancer treatment.This article provides a review of the effects of HIF-1 on cellular glucose metabolism,tumor metastasis,and targeted therapy in digestive tract tumor igenesis.
