摘要
课题组前期试验证明黑木耳九丰为优良的富硒发酵菌种。该试验在前期研究基础上,通过单因素及响应面试验优化硒蛋白提取工艺,并对提取出的硒蛋白进行硒含量测定、体外抗肿瘤试验以及抗氧化活性研究。结果表明,硒蛋白最适提取条件为碱液浓度0.077 mol/L、超声时间1.404 h、料液比1:93.387 g/mL,此条件下提取率为19.81%。同时,硒蛋白中硒含量达到了197.07μg/g,并且对结直肠癌HCT116细胞具有一定的抑制作用,当硒蛋白样品浓度为1000μg/mL,上药时间24 h时,对结直肠癌细胞的抑制率达到了51.07%。以硒蛋白清除1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基、羟自由基和总还原力为指标,发现其具有较强的体外抗氧化活性。研究结果为进一步探究富硒黑木耳菌丝体蛋白的提取方法和开发具有抗肿瘤、抗氧化功效的产品提供了依据,可为富硒黑木耳资源的综合利用提供参考。
The research group’s previous experiments proved that Auricularia heimuer Jiufeng is an excellent selenium-rich fermentation strain.On the basis of previous studies,single factor and response surface tests were used to optimize the extraction process of selenoprotein,and selenium content,in vitro anti-tumor test and antioxidant activity of the extracted selenoprotein were determined.The results showed that the optimal extraction conditions were as follows:lye concentration 0.077 mol/L,ultrasonic time 1.404 h,solid-liquid ratio 1:93.387 g/mL.The extraction rate was 19.81%under these conditions.At the same time,the selenium content of selenoprotein reached 197.07μg/g,and had a certain inhibitory effect on colorectal cancer HCT116 cells.When the concentration of selenoprotein sample was 1000μg/mL and the time of administration was 24 h,the inhibitory rate of colorectal cancer cells reached 51.07%.Selenium protein scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical,hydroxyl free radical and total reducing power were used as indexes.It was found that selenium protein had strong antioxidant activity in vitro.The results provide a basis for further exploration of the extraction method of selenium-rich Auricularia heimuer mycelia protein and the development of products with anti-tumor and anti-oxidation effects,and can provide a reference for the comprehensive utilization of selenium-rich Auricularia heimuer resources.
