响应面法优化箭叶淫羊藿蛋白提取工艺研究

Optimization of the Epimedium sagittatum Protein Extraction Process Using Response Surface Methodology

摘要 目的优化箭叶淫羊藿蛋白的提取工艺并测定原药材及提取物中总蛋白含量。方法以蛋白提取率为指标筛选出最佳蛋白质提取方法;在单因素实验基础上,利用Box-Behnken响应面法优化淫羊藿蛋白提取工艺,并测定出淫羊藿蛋白等电点,最后通过凯士定氮法对淫羊藿原药材和提取物中的蛋白质进行定量检测。结果淫羊藿蛋白碱提法最优提取工艺参数:提取温度为35℃、溶液pH为11、提取时间为1.5 h、液料比1∶20(g/mL),该条件下提取率达到5.06±0.03%。同时,通过凯士定氮法得出淫羊藿粉末的蛋白质含量为122.35±1.94 g/kg,提取物中蛋白质含量为258.85±1.53 g/kg。结论实验表明,优化得到的淫羊藿蛋白提取工艺稳定可行,可以用于工业化生产。 Objective To optimize the protein extraction process from Epimedium sagittatum and to quantify the total protein content in both raw herb material and extracts.Methods The optimal protein extraction method was initially screened based on extraction yield as the evaluation index.Single-factor experiments were conducted,followed by process optimization using the Box-Behnken design of response surface methodology(RSM).The isoelectric point of Epimedium protein was determined,and final protein quantification in both raw materials and extracts was performed using the Kjeldahl method.Results The optimal extraction conditions were a temperature of 35,a solution pH of 11,an extraction time of 1.5 hours,and a liquid-to-solid ratio of 1:20(g/mL).Under these conditions,the protein yield reached 5.06±0.03%.Kjeldahl analysis showed that the protein content was 122.35±1.94 g/kg in the raw herb powder and 258.85±1.53 g/kg in the extract.Conclusion The experiments showed that the optimised Epimedium protein extraction process was stable and feasible,and could be used for industrial production.
