基于AHP-熵权法结合正交设计优选箭叶淫羊藿汤剂工艺

Optimization of the Extraction Process of Epimedium sagittatum Decoction Based on the Combination of AHP-Entropy Weight Method and Orthogonal Design

摘要 目的建立箭叶淫羊藿汤剂中多个成分的含量测定方法,优化箭叶淫羊藿汤剂煎煮工艺,为后续制备配方颗粒提供一定科学依据。方法通过测定箭叶淫羊藿汤剂中指标成分含量,在单因素实验的基础上采用正交试验法考察煎煮时间、煎煮次数、加水倍数等影响因素,以箭叶淫羊藿汤剂中4种特征性黄酮类成分含量、总黄酮含量及出膏率作为指标,采用层次分析法结合熵权法对各指标进行赋权,计算综合评分进行工艺优选。结果箭叶淫羊藿煎煮最佳工艺为浸泡30 min,煎煮三次,每次煎煮60 min、40 min、40 min,分别加水32倍、30倍、30倍。结论建立的多成分含量测定方法操作便捷准确,优选的煎煮工艺简便、稳定可行,为箭叶淫羊藿工业化生产提供一定科学依据。 Objective To establish a method for the determination of muliple components in Epimedium sagitatum decoction,and to optimize the decoction process,providing a scientific basis for the subsequent preparation of formula granules.Methods Based on the measurement of characteristic components in the decoction,an orthogonal experimental design was conducted following single-factor experiments to investigate the effects of boiling time,number of boilings,and water-to-material ratio.The contents of four characteristic flavonoid compounds,total flavonoid content,and extract yield were used as evaluation indices.The analytic hierarchy process(AHP)combined with the entropy weight method was applied to assign weights to each indicator,and a comprehensive score was calculated to identify the optimal process.ResultsThe optimal decoction conditions were soaking for 30 minutes,boiling three times for 60 minutes,40 minutes,and 40 minutes respectively,with water added at 32-fold,30-fold,and 30-fold the material weight for each decoction.Conclusion The established multi-component HPLC determination method is convenient and accurate.The optimized decoction process is simple,stable,and feasible,providing a scientific foundation for the industrial production of Epimedium sagittatum.
