摘要
以梅州沙田柚籽为原料,采用亚临界法萃取柚籽油,乙酸法提取其中柠檬苦素类化合物,探索最佳提取工艺条件,并通过高效液相色谱、半制备液相色谱以及核磁共振谱分析富集并鉴定柚籽油中的柠檬苦素类化合物。结果表明,最佳提取工艺为提取温度54℃、提取时间2.9 h、柚籽油与乙酸体积比1∶5.2、乙酸浓度80.3%,柚籽油中黄柏酮提取量达64.34μg/g。沙田柚籽油中共鉴定出4种柠檬苦素类化合物,分别为黄柏酮、柠檬苦素、诺米林及另一未知化合物。该未知化合物通过核磁共振技术以及质谱技术鉴定为诺米林酸。
Meizhou Shatian pomelos were used as raw materials for extraction of pomelo seed oil by the subcritical method,and limonoids were extracted by acetic acid.The optimal extraction conditions were explored,and the limonoids in the pomelo seed oil were identified and enriched by high performance liquid chromatography,semi-preparative liquid chromatography,and nuclear magnetic resonance spectrometry.The results showed that the optimal extraction conditions were 54℃,2.9 h,pomelo seed oil∶acetic acid ratio of 1∶5.2(mL/mL),and 80.3%acetic acid,under which the yield of obacunone extracted from pomelo seed oil reached as high as 64.34μg/g.Four limonoids were identified in the Shatian pomelo seed oil,including obacunone,limonin,nomilin,and an unknown compound.The unknown compound was identified as nomilinic acid by nuclear magnetic resonance and mass spectrometry.
