氟西汀改善慢性不可预见温和应激诱导的猪抑郁样状态

Protective Effect of Fluoxetine on Depressive-like State in Pigs after Chronic Unforeseeable Mild Stress

摘要 在生猪的集约化养殖中,环境贫瘠、空间狭窄等因素导致的慢性应激对猪的行为和生理造成严重负面影响,使其产生类抑郁状态,进而影响猪的福利。为了探讨可行的改善猪抑郁样状态的方法,研究将经过慢性不可预见温和应激(CUMS)造模的16头成年巴马香猪随机分为氟西汀处理组(n=8)和自然恢复处理组(n=8),对氟西汀处理组进行4周的氟西汀给药,自然恢复处理组不进行任何干预。对各处理组初始状态、造模后0、2、4周进行行为评估、生理指标检测以及海马组织miRNA表达量测定。结果表明:氟西汀提高造模后2周和4周猪的糖水偏好率,降低了接触新事物的潜伏期,提高了造模后4周的新事物接触次数和接触时间。同时,氟西汀还可以降低猪造模后4周血清中促肾上腺皮质激素释放激素(COR)含量,但对于血清促肾上腺皮质激素(ACTH)含量没有影响,降低造模后4周猪海马体中miR-34a的相对表达量,提高miR-16、miR-124的相对表达量。综上所述,经氟西汀干预可以有效改善CUMS猪的抑郁样行为,降低血液中COR含量,调节海马体中miRNA表达,提高集约化饲养过程中猪的福利水平。 In the intensive production of pigs,the chronic stress caused by the barren environment,narrow space and other factors has a serious negative impact on the behavior and physiology of pigs,causing them to produce a depression-like state,and then affecting the welfare of pigs.In order to explore a feasible method to improve depressive-like state of pigs,16 adult Bamea pigs modeled by Chronic Unforeseeable Mild Stress(CUMS)were randomly divided into fluoxetine treatment group(n=8)and natural recovery treatment group(n=8).The fluoxetine treatment group was given fluoxetine for 4 weeks and without any intervention in the natural recovery treatment group.The results showed that fluoxetine increased the sugar water preference rate of pigs at 2 and 4 weeks after modeling,reduced the incubation period of exposure to novelty stimulus,and increased the number and time of exposure to novelty stimulus at 4 weeks after modeling.At the same time,fluoxetine could also reduce the serum corticotropin releasing hormone(COR)content of pigs at 4 weeks after modeling,but had no effect on the serum adrenocorticotrophic hormone(ACTH)content.It reduced the relative expression level of miR-34a,increased the relative expression levels of miR-16 and miR-124 in the hippocampus of pigs at 4 weeks after modeling.In conclusion,fluoxetine intervention could effectively improve the depression-like behavior of CUMS pigs,reduce the content of COR in blood,regulate the expression of miRNA in the hippocampus,and improve the welfare level of pigs in the intensive feeding process.
