番茄CBF基因家族全基因组分析

摘要 为鉴定番茄CBF基因家族、初步探究SlCBFs基因的生物学功能,基于拟南芥、水稻和马铃薯CBF基因,在番茄(Solanum lycopersicum L.)中筛选出6个CBF基因,并对其理化特征和组织表达模式等进行了系统的比较分析,明确了CBF基因家族在番茄响应低温胁迫的功能。结果表明:1)系统进化树分析显示植物CBF基因家族分为4个亚组,其中番茄SlCBFs家族成员分布在GroupⅠ和GroupⅡ2个亚组中,与水稻CBF基因家族所在的GroupⅣ存在明显区别;2)顺式作用元件分析发现了1072个可能调控CBF基因表达的顺式元件,其中生长发育响应元件最多,占72%;3)组织转录表达分析结果表明,SlCBF1、SlCBF2、SlCBF3和SlCBF4基因在果实破色期表达量较高,推断SlCBFs基因家族在植物果实发育过程中可能起到重要作用;4)低温胁迫处理发现SlCBF1、SlCBF2、SlCBF3、SlCBF4和SlCBF5基因表达水平明显上调。综上,深入探究番茄CBF基因家族的生物学功能,可为番茄新品种创制提供理论基础。 To identify tomato CBF gene family and the preliminary exploration of the biological function of the SlCBFs gene,therefore,in this research,six CBF genes in tomato(Solanum lycopersicum L.)were identified based on the CBF genes of Arabidopsis,rice,and potato.The physicochemical characteristics and tissue expression patterns were systematically compared and analyzed to clarify the function of the CBF gene family in tomato.The results are as follows:1)The phylogenetic tree analysis indicated that all CBF genes were divided into four subgroups.The SlCBFs gene family members of tomato were distributed in two subgroups,GroupⅠand GroupⅡ,which were significantly different from the CBF gene of rice(GroupⅣ).2)The analysis of cis-acting elements revealed a total of 1072 cis-acting elements could regulate the expression of the CBF gene,among which the growth and development elements were the most,accounting for 72%.3)Tissue transcription analysis demonstrated that SlCBF1~SlCBF4 were expressed at relatively high levels during the color breaking stage,suggesting that the SlCBFs gene might play a significant role in plant growth and development.4)The expression levels of SlCBF1~SlCBF5 were significantly up-regulated under low-temperature stress.To conclude,the biological function of the tomato CBF gene family and the mechanism of tomato cold tolerance can provide a theoretical basis for new tomato variety creation.
