摘要
目的探讨垂体瘤组织中微小RNA-103a-3p(miR-103a-3p)和高迁移率组蛋白1(HMGB1)的表达水平,及其在垂体瘤中的临床意义。方法研究对象为2017年4月至2019年4月神经外科和内分泌科收治的垂体瘤患者73例,在手术过程中收集垂体瘤组织和正常垂体组织。应用实时荧光定量PCR法检测各组织中miR-103a-3p和HMGB1mRNA的相对表达水平,Western blot法检测各组织中HMGB1蛋白的相对表达量,比较miR-103a-3p和HMGB1在各组织中表达水平的差异,分析垂体瘤组织中miR-103a-3p、HMGB1蛋白表达与垂体瘤临床病理特征的关系,使用TargetScan网站预测HMGB1与miR-103a-3p的靶向关系,使用Pearson相关性分析miR-103a-3p、HMGB1蛋白表达在垂体组织中的关系。结果垂体瘤组织中,miR-103a-3p表达下调(P<0.05),HMGB1 mRNA和蛋白表达水平均上调(P<0.05)。垂体瘤组织中miR-103a-3p低表达和HMGB1蛋白高表达均与垂体瘤侵袭性有关(P<0.05);miR-103a-3p和HMGB1具有靶向关系,其中miR-103a-3p与HMGB1蛋白在垂体瘤组织中的表达呈负相关(r=-0.806,P<0.05)。结论miR-103a-3p在垂体瘤发病机制中具有抑癌作用,HMGB1具有致癌作用,两者表达关系密切,均与垂体瘤侵袭性有关,可能作为临床诊断垂体瘤发生发展的标志物。
Aim To detect the expression levels of microRNA-103a-3p(miR-103a-3p)and high mobility group box 1(HMGB1)in pituitary adenoma tissues,and discuss their clinical significance in patients with pituitary adenoma.Methods This study included 73 pituitary adenoma patients admitted to the department of neurosurgery and endocrinology from April 2017 to April 2019.Pituitary adenoma tissue and normal pituitary tissue were collected during the operation.Real-time fluorescence quantitative PCR was used to detect the relative expression levels of miR-103a-3p and HMGB1 mRNA in various tissues,Western blot method was used to detect the relative expression level of HMGB1 protein in each tissue,t-test was used to compare the expression levels of miR-103a-3p and HMGB1 in various tissues,χ2 test was used to analyze the relationship between the expression of miR-103a-3p and HMGB1 protein in pituitary adenoma tissue and the clinicopathological characteristics of patients with pituitary adenoma,the Target Scan website was used to predict the targeting relationship between HMGB1 and miR-103a-3p,Pearson correlation was used to analyze the relationship between miR-103a-3p and HMGB1 protein expression in pituitary tissues.Results The expression of miR-103a-3p was down-regulated in pituitary adenoma tissues(P<0.05),and the expression levels of HMGB1 mRNA and protein were up-regulated in pituitary adenoma tissues(P<0.05).Both the low expression of miR-103a-3p and the high expression of HMGB1 protein in pituitary adenoma tissue were related to the invasion of pituitary adenoma patients(P<0.05).There was a targeting relationship between miR-103a-3p and HMGB1,and the expression of miR-103a-3p and HMGB1 protein in pituitary adenoma tissue was negatively correlated(r=-0.806,P<0.05).Conclusion MiR-103a-3p plays a tumor suppressor effect in the pathogenesis of pituitary adenoma,and HMGB1 plays a carcinogenic effect.The expression of the two is closely related,and both are related to the invasion of pituitary adenoma,and may be used as markers for clinical diagnosis of the occurrence and development of pituitary adenoma.
