基于“肠-皮”轴研究银屑平丸对银屑病样小鼠模型短链脂肪酸的影响

摘要 目的:探讨银屑平丸通过调控“肠-皮”轴对咪喹莫特诱导的银屑病样小鼠模型短链脂肪酸含量的影响。方法:将60只BALB/c小鼠随机分为对照组、模型组、银屑平丸高、中、低剂量组及抗生素组,每组各10只。给药8天后,比较各组小鼠PASI评分;HE染色观察皮损组织形态学改变;液相色谱-质谱法检测结肠内容物中SCFAs含量。结果:除对照组外,其余各组小鼠造模后背部皮损PASI评分均升高(P﹤0.05);与模型组比较,银屑平丸高、中、低剂量组PASI评分均有一定降低(P﹤0.05),抗生素组小鼠PASI评分无明显改变(P﹥0.05)。与对照组比较,模型组小鼠皮损组织呈现典型的银屑病角化过度伴角化不全改变,而银屑平丸高、中、低剂量组变化均较少。与对照组比较,模型组SCFAs表达量明显下降;与模型组比较,银屑平丸高、中、低剂量组SCFAs表达量均有所上升,中剂量组上升趋势较为明显(P﹤0.05);抗生素组SCFAs表达量较模型组进一步降低。结论:银屑平丸能降低银屑病样小鼠皮损PASI评分,可能通过调控“肠-皮”轴促进小鼠SCFAs的生成,从而改善、治疗银屑病皮损。 Objective:To approach the effect of Yinxieping pill on the content of short-chain fatty acids in imiquimod-induced modeled mice with psoriasis-like by regulating"intestine-skin"axis.Methods:60 BALB/c mice were randomly divided into control group,model group,high-,middle-and low-dose groups of Yinxieping pill and antibiotic group,10 mice in each group.PASI scores of mice were compared in each group after 8 days of administration.The histomorphological changes of skin lesion was observed by HE staining.SCFAs content in colon content was detected by liquid chromatography-mass spectrometry.Results:Except for the control group,PASI scores of skin lesions on back of mice were increased in other groups after modeling(P﹤0.05).Compared with the model group,PASI scores of high-,middle-and low-dose groups of Yinxieping pill were decreased to some extent(P﹤0.05);while PASI scores of mice in antibiotic group were changed unsignificantly(P﹥0.05).Compared with the control group,the histomorphology of skin lesion of mice in model group showed typical psoriasis hyperkeratosis with parakeratosis change,while the changes were less in high-,middle-and low-dose groups of Yinxieping pill.Compared with the control group,SCFAs expression of model group was decreased significantly.Compared with the model group,SCFAs expressions of high-,middle-and low-dose groups of Yinxieping pill were increased,and the increasing trend of middle-dose group was more obvious(P﹤0.05).SCFAs expression of antibiotic group was further decreased compared with model group.Conclusion:Yinxieping pill can reduce PASI score of skin lesion of mice with psoriasis-like,and may promote the generation of SCFAs in mice by regulating"intestine-skin"axis,in order to improve and treat psoriasis-induced lesions.
