摘要
本文研究防御酶的代谢反应及抗病相关基因表达与棉花抗黄萎病之间的关系,为棉花抗黄萎病材料的选择提供了依据。田间病圃研究发现,中植棉2号和新陆早36号在6~7片真叶时开始感病,在花期,新陆早36号发病率和病情指数显著高于中植棉2号,表现为高感材料,而中植棉2号则为高抗材料。生理生化分析显示,中植棉2号中CHI含量整体高于新陆早36号品种,POD和PPO在处理24 h和120 h含量高于新陆早36号,PAL在处理120 h时含量高于新陆早36号;进一步分析了抗病相关基因的表达特征,结果显示,中植棉2号中CHI、AOS、ACO基因的表达量在处理时间点高于新陆早36号,AOC基因的表达量在处理时间点24、72、120 h高于新陆早36号,PR1基因的表达量在处理时间点120、168 h高于新陆早36号,EDS1基因的表达量在处理时间点168 h高于新陆早36号。通过田间病圃进行材料鉴定,并结合不同抗性品种接种黄萎病菌后检测防御酶活性的变化和抗性基因的表达,有助于棉花抗黄萎病种质的评估。
The study of the metabolic reactions of defense enzymes and the expression patterns of disease resistance-related genes in relation to cotton's resistance to Verticillium wilt lays the groundwork for selecting cotton varieties with enhanced disease resistance.Field disease nursery trials have revealed that Zhong Zhi Mian 2 and Xinlu Zao 36 begin to exhibit symptoms at the stage of 6 to 7 true leaves.Notably,during the flowering period,Xinlu Zao 36 shows a significantly higher incidence and disease index compared to Zhong Zhi Mian 2 classifying it as a highly susceptible variety,whileZhong Zhi Mian 2 is identified as highly resistant.Physiological and biochemical analyses indicate that Zhong Zhi Mian 2 has a higher overall content of CHI compared to Xinlu Zao 36.Additionally,the levels of POD and PPO are elevated in Zhong Zhi Mian 2 at 24 and 120 hours post-treatment,and the PAL content surpasses that of Xinlu Zao 36 at the 120-hour mark post-treatment.Further examination of the expression profiles of disease resistance-related genes reveals that the expression levels of CHI,AOS,and ACO genes in Zhong Zhi Mian 2 are higher than those in Xinlu Zao 36 at the respective treatment time points.The AOC gene shows increased expression at 24,72,and 120 hours post-treatment,the PR1 gene at 120 and 168 hours,and the EDS1 gene at 168 hours,all in Zhong Zhi Mian 2 compared to Xinlu Zao 36.Conducting material identification through field disease nurseries,and combining the detection of changes in defense enzyme activity and the expression of resistance genes after inoculating different resistant varieties with Verticillium wilt,aids in the assessment of cotton germplasm resistant to Verticillium wilt.
